目的:评价16种与阿尔茨海默病相关的天然药物提取物对β淀粉样蛋白( Aβ42)寡聚体致PC-12细胞毒性的作用。方法通过超声波破碎法获得16种天然药物的提取物,并筛选具有抑制Aβ42寡聚体诱导的神经毒性作用的药物。将PC-12细胞随机分为实验组、细胞损伤对照组、天然药物对照组、正常细胞对照组。通过细胞培养液中加入Aβ42寡聚体诱导建立PC-12细胞损伤模型。采用MTT比色法测定各组细胞存活率。结果苦参、石菖蒲、何首乌、远志、当归、女贞子、黄芪、益智仁、绞股蓝和淫羊藿10种天然药物提取物具有抑制Aβ42寡聚体诱导神经毒性的作用,其中苦参碱的提取物抑制作用最强。细胞损伤对照组PC-12细胞的存活率显著高于实验组、天然药物对照组及正常细胞对照组( P均<0.05)。结论最终确定10种天然药物(苦参、石菖蒲、何首乌、远志、当归、女贞子、黄芪、益智仁、绞股蓝和淫羊藿)提取物具有抑制Aβ42寡聚体诱导的神经毒性作用,苦参碱的提取物抑制作用最强。

목적:평개16충여아이자해묵병상관적천연약물제취물대β정분양단백( Aβ42)과취체치PC-12세포독성적작용。방법통과초성파파쇄법획득16충천연약물적제취물,병사선구유억제Aβ42과취체유도적신경독성작용적약물。장PC-12세포수궤분위실험조、세포손상대조조、천연약물대조조、정상세포대조조。통과세포배양액중가입Aβ42과취체유도건립PC-12세포손상모형。채용MTT비색법측정각조세포존활솔。결과고삼、석창포、하수오、원지、당귀、녀정자、황기、익지인、교고람화음양곽10충천연약물제취물구유억제Aβ42과취체유도신경독성적작용,기중고삼감적제취물억제작용최강。세포손상대조조PC-12세포적존활솔현저고우실험조、천연약물대조조급정상세포대조조( P균<0.05)。결론최종학정10충천연약물(고삼、석창포、하수오、원지、당귀、녀정자、황기、익지인、교고람화음양곽)제취물구유억제Aβ42과취체유도적신경독성작용,고삼감적제취물억제작용최강。
Objective To evaluate the effects of 16 extracts of Alzheimer disease-related natural Chinese medicine on Aβ42 oligomer induced PC-12 cell toxicity .Methods The effective components of 16 natural Chinese medicines were ex-tracted by ultrasonic method and the extracts which had the effect of inhibiting the cytotoxicity of A β42 were sreened .The PC-12 cells were randomly divided into the experimental group , cell damage group , natural medicine control group and nor-mal cells control group .The extracts were added to the culture medium of the PC 12 cell models respectively which was in-duced by the Aβ42 oligomers, MTT colorimetric analysis was used to determine the cell survival rate .Results Matrine, Shichangpu , Radix Polygoni Multiflori , Radix Polygalae , Chinese angelica , Ligustrum lucidum , astragalus root , Fructus alpiniae oxyphyllae , Gynostemma pentaphyllum and Epimedium , a total of 10 extracts had the effect of inhibiting the cyto-toxicity of Aβ42 .The extract of Matrine had the strongest effect .The survival rate in PC-12 cells of the cell damage group was significantly higher than that of the experimental group , natural medicine control group and normal cells control group (all P<0.05).Conclusion We finally identify 10 extracts (Matrine, Shichangpu, Radix Polygoni Multiflori, Radix Po-lygalae, Chinese angelica , Ligustrum lucidum , astragalus root , Fructus alpiniae oxyphyllae , Gynostemma pentaphyllum and Epimedium) have the effect of inhibiting the cytotoxicity of Aβ42, and Matrine extract has the strongest effect .
