目的:观察黄芪多糖(APS)对四氯化碳(CCl4)致大鼠肝纤维化的治疗作用。方法:采用 CCl4诱导大鼠肝纤维化模型,造模成功后,取大鼠血清和肝组织进行检测。测定血清肝纤维化四项、肝脏组织中羟脯氨酸(HYP)含量;HE 染色比较炎症及肝纤维化程度;免疫组织化学法分析肝组织中 I 型胶原(Col -I)、平滑肌肌动蛋白α(α-SMA)表达量。结果:与模型组相比,APS 中、高剂量组的肝纤四项水平、炎症及肝纤维化程度明显降低(P <0.05);APS 中、高剂量组肝脏组织中HYP 含量显著降低(P <0.01);APS 中剂量组 Col -I、α-SMA 表达显著减少(P <0.01);APS 高剂量组 Col -I、α-SMA表达明显减少(P <0.05)。结论:APS 对大鼠肝纤维化具有一定的防治作用,作用机制可能与其抑制 Col -I、α-SMA 表达有关。

목적:관찰황기다당(APS)대사록화탄(CCl4)치대서간섬유화적치료작용。방법:채용 CCl4유도대서간섬유화모형,조모성공후,취대서혈청화간조직진행검측。측정혈청간섬유화사항、간장조직중간포안산(HYP)함량;HE 염색비교염증급간섬유화정도;면역조직화학법분석간조직중 I 형효원(Col -I)、평활기기동단백α(α-SMA)표체량。결과:여모형조상비,APS 중、고제량조적간섬사항수평、염증급간섬유화정도명현강저(P <0.05);APS 중、고제량조간장조직중HYP 함량현저강저(P <0.01);APS 중제량조 Col -I、α-SMA 표체현저감소(P <0.01);APS 고제량조 Col -I、α-SMA표체명현감소(P <0.05)。결론:APS 대대서간섬유화구유일정적방치작용,작용궤제가능여기억제 Col -I、α-SMA 표체유관。
Objective:To study the protective effect of the Astragalus polysaccharides (APS)on liver fibrosis in rats caused by carbon tetrachloride (CCl4 ).Methods:CCl4 was used to induce rat liver fibrosis model.After making the model successfully,rat serum and liver tissue were collected to test.The serum liver fiber four and hydroxyproline (HYP)content in the liver tissue was tested;HE staining of liver tissue was used to compare inflammation and degree of liver fibrosis;expression of Col-I and α-SMA with immunohistochemical method was analyzed.Results:Compared with the model group,four levels of liver fibrosis,inflamma-tion and liver fibrosis of APS high and medium groups were obviously lower(P <0.05);HYP content of APS high and medium groups were significantly lower (P <0.01 );Col-I and α-SMA expression of APS medium group was significantly lower (P <0.01 );Col-I and α-SMA expression of APS high group was obviously lower (P <0.05).Conclusion:The APS is effective to some extent in preventing liver fibrosis of rat,the functional mechanism may be related to limiting the expression of Col-I and α-SMA.
