目的:通过检测PDCD4和DNMTI在胃癌组织、癌旁组织、正常组织中的表达,研究其临床意义。方法选取2012年6月至2013年3月9个月间在我院进行治疗的胃癌患者,采用免疫组织化学SP法检测60例胃癌组织、60例癌旁组织、60例正常胃黏膜组织。收集三组患者PDCD4和DNMTI的表达水平、阳性率,用RT-PCR法检测PDCD4和DNMTImRNA的表达,研究其病理学参数关系,并对收集的数据进行统计学分析。结果 DNMTI在胃癌组织、癌旁组织、正常组织中阳性含量为71.67%、61.67%、36.67%,PDCD4在胃癌组织、癌旁组织、正常组织中阳性含量为30%、50%、70%,表现出相反的趋势,经表达相关性分析成负相关关系。胃癌组织与癌旁组织比较数据差异不明显,不具有统计学意义(P>0.05)。而胃癌组织、癌旁组织与正常组织相比较数据差异明显,具有统计学意义(P<0.05)。 PDCD4和DNMTI的表达都与病理分型存在显著相关性(P<0.05)。结论 PDCD4和DNMTI的表达出现异常与胃癌的发生、发展存在密切联系,两者表达成负相关关系。

목적:통과검측PDCD4화DNMTI재위암조직、암방조직、정상조직중적표체,연구기림상의의。방법선취2012년6월지2013년3월9개월간재아원진행치료적위암환자,채용면역조직화학SP법검측60례위암조직、60례암방조직、60례정상위점막조직。수집삼조환자PDCD4화DNMTI적표체수평、양성솔,용RT-PCR법검측PDCD4화DNMTImRNA적표체,연구기병이학삼수관계,병대수집적수거진행통계학분석。결과 DNMTI재위암조직、암방조직、정상조직중양성함량위71.67%、61.67%、36.67%,PDCD4재위암조직、암방조직、정상조직중양성함량위30%、50%、70%,표현출상반적추세,경표체상관성분석성부상관관계。위암조직여암방조직비교수거차이불명현,불구유통계학의의(P>0.05)。이위암조직、암방조직여정상조직상비교수거차이명현,구유통계학의의(P<0.05)。 PDCD4화DNMTI적표체도여병리분형존재현저상관성(P<0.05)。결론 PDCD4화DNMTI적표체출현이상여위암적발생、발전존재밀절련계,량자표체성부상관관계。
Objective To detect the expression of DNMTI and PDCD4 in gastric cancer tissues, adjacent tissues and normal tissues, and study its clinical significance. Methods The gastric cancer patients who treated in our hospital from June 2012 to March 2013 were Selected. The immunohisto-chemistry SP method utilized to detecte 60 cases of gastric carcinoma tissues, can-cer adjacent tissues and normal gastric mucosal tissues, respectively. was The expression level and positive rate of PDCD4 and DNMT1 among three groups were analyzed. The expression of PDCD4 and DNMTI mRNA was determined by RT-PCR assay. The relationship of its pathological parameters were statistically analyzed. Results The positive content of DNMTI in gastric cancer tis-sues, adjacent tissues and normal tissues was 71.67%, 61.67%, 36.67%, respectively; the positive content of PDCD4 was 30%, 50%, 70%, respectively, which showed an opposite trend and persented negative correlation by expressed correlation analysis. The data of gastric cancer tissues compared with adjacent tissues has no significantly difference (P>0.05). However, the data of gastric cancer tissues andadjacent tissues compared with normal tissues has significantly difference(P<0.05). The expression of PDCD4 and DNMTI significantly association with pathological typing(P<0.05). Conclusion Abnormal expression of PDCD4 and DNMTI closely related to the occurrence and development of gastric cancer, andshowed a negative correlation.
