目的:获得融合表达的Her2/ECD-sf162/TM杆状病毒,为后续基于SIV Gag蛋白的Her2病毒样颗粒疫苗的制备奠定实验基础。方法:将Her2胞外段基因与猴免疫缺陷病毒( simian immunodeficiency virus, SIV)包膜蛋白sf162跨膜区基因融合的Her2/ECD-sf162/TM PFastBac重组表达载体,转座大肠杆菌E. coli DH10Bac菌株,获得重组杆粒,在昆虫细胞中表达获得重组杆状病毒,Western blot方法对该病毒进行鉴定。结果:构建的Her2/ECD-sf162/TM杆状病毒表达载体经PCR鉴定正确,转染昆虫细胞后获得了重组杆状病毒,该融合杆状病毒蛋白可与抗Her2抗体发生免疫反应。结论:成功构建的Her2/ECD-sf162/TM杆状病毒可用于Her2病毒样颗粒疫苗的制备。
目的:穫得融閤錶達的Her2/ECD-sf162/TM桿狀病毒,為後續基于SIV Gag蛋白的Her2病毒樣顆粒疫苗的製備奠定實驗基礎。方法:將Her2胞外段基因與猴免疫缺陷病毒( simian immunodeficiency virus, SIV)包膜蛋白sf162跨膜區基因融閤的Her2/ECD-sf162/TM PFastBac重組錶達載體,轉座大腸桿菌E. coli DH10Bac菌株,穫得重組桿粒,在昆蟲細胞中錶達穫得重組桿狀病毒,Western blot方法對該病毒進行鑒定。結果:構建的Her2/ECD-sf162/TM桿狀病毒錶達載體經PCR鑒定正確,轉染昆蟲細胞後穫得瞭重組桿狀病毒,該融閤桿狀病毒蛋白可與抗Her2抗體髮生免疫反應。結論:成功構建的Her2/ECD-sf162/TM桿狀病毒可用于Her2病毒樣顆粒疫苗的製備。
목적:획득융합표체적Her2/ECD-sf162/TM간상병독,위후속기우SIV Gag단백적Her2병독양과립역묘적제비전정실험기출。방법:장Her2포외단기인여후면역결함병독( simian immunodeficiency virus, SIV)포막단백sf162과막구기인융합적Her2/ECD-sf162/TM PFastBac중조표체재체,전좌대장간균E. coli DH10Bac균주,획득중조간립,재곤충세포중표체획득중조간상병독,Western blot방법대해병독진행감정。결과:구건적Her2/ECD-sf162/TM간상병독표체재체경PCR감정정학,전염곤충세포후획득료중조간상병독,해융합간상병독단백가여항Her2항체발생면역반응。결론:성공구건적Her2/ECD-sf162/TM간상병독가용우Her2병독양과립역묘적제비。
Objective:To gain Her2/ECD-sf162/TM recombinant baculovirus,laiding a foundation for further study of Her2 virus like Particles vaccine that based on SIV Gag Protein. Methods:Her2/ECD -sf162/TM fusion gene recombinant PFastBac-1 exPression vector of Her2 extracellular domain and SIV enveloPe Protein sf162 trans-membrane domain was constructed,and then transformed into E. coli strain DH10Bac to gain recombinant bacmids, which then were transfected into insect cells to exPress recombinant baculovirus with bac-to-bac systerm. In turn, Western blot were Performed identificated the recombination. Results:Recombinant PFastBac -1 exPression vector was successfully constructed and high titer recombinant baculovirus which could imunoreacted with Her2 antibody were gained. Conclusion:Her2/ECD -sf162/TM recombinant baculovirus can be used to develoP Her2 virus like Particles vaccine.
