目的:探讨柴胡对自身免疫性脑脊髓炎(EAE)小鼠干预作用,为临床应用柴胡治疗多发性硬化(MS)提供依据。方法C57雌性小鼠40只,随机分为空白对照组、EAE模型组、泼尼松组[7.8 mg/(kg· d)]和柴胡组[1.95 g/(kg· d)],每组10只。EAE模型组、泼尼松组和柴胡组6周龄时用 MOG3555多肽免疫建立 EAE模型,造模后第3天灌胃给予蒸馏水或药物至实验结束。自造模当天观察各组小鼠体重、行为学;造模后第16天(发病高峰期)取脊髓组织行 HE染色,观察炎性细胞浸润情况;采取尾静脉血,检测与多发性硬化主要相关炎症因子白细胞介素(IL)17E、γ干扰素(IFN γ)、IL 4、IL 21、IL 10、IL 23、IL 12(p70)、IL 17A、IL 17F 和肿瘤坏死因子(TNF)α水平。结果与空白对照组比较,EAE模型组小鼠发病高峰外周血中 IL 17E、IFN γ、IL 17F水平升高(P<0.05),IL 4水平降低(P<0.05);与 EAE模型组比较,发病高峰期泼尼松组外周血中 IL 17E、IFN γ、IL 17F水平降低(P<0.05或P<0.01),柴胡组 IL 17E和 IFN γ水平降低(P<0.05或P<0.01)。结论柴胡通过抗炎机制减轻 EAE小鼠的病情,可作为 MS治疗的参考。

목적:탐토시호대자신면역성뇌척수염(EAE)소서간예작용,위림상응용시호치료다발성경화(MS)제공의거。방법C57자성소서40지,수궤분위공백대조조、EAE모형조、발니송조[7.8 mg/(kg· d)]화시호조[1.95 g/(kg· d)],매조10지。EAE모형조、발니송조화시호조6주령시용 MOG3555다태면역건립 EAE모형,조모후제3천관위급여증류수혹약물지실험결속。자조모당천관찰각조소서체중、행위학;조모후제16천(발병고봉기)취척수조직행 HE염색,관찰염성세포침윤정황;채취미정맥혈,검측여다발성경화주요상관염증인자백세포개소(IL)17E、γ간우소(IFN γ)、IL 4、IL 21、IL 10、IL 23、IL 12(p70)、IL 17A、IL 17F 화종류배사인자(TNF)α수평。결과여공백대조조비교,EAE모형조소서발병고봉외주혈중 IL 17E、IFN γ、IL 17F수평승고(P<0.05),IL 4수평강저(P<0.05);여 EAE모형조비교,발병고봉기발니송조외주혈중 IL 17E、IFN γ、IL 17F수평강저(P<0.05혹P<0.01),시호조 IL 17E화 IFN γ수평강저(P<0.05혹P<0.01)。결론시호통과항염궤제감경 EAE소서적병정,가작위 MS치료적삼고。
Objective To explored the effect of Chaihu for treatment of experimental autoimmune encephalomyelitis(EAE),and to pro-vide laboratory evidence for clinical application of Chaihu treating multiple sclerosis.Methods C57 mice were immunized by MOG 35 55 to establish EAE,the model of multiple sclerosis.Forty mice were divided into blank control group,EAE model group,predni-sone treatment group and Chaihu treatment group,which received oral administrations of different drugs after immunized 3 days.The mice behavior,checked daily and graded on clinical score were observed.It had been observed that cytokines levels of peripheral blood,relating to multiple sclerosis major factors,and inflammatory cel infiltration in spinal by HE staining after immunized 16 days. Results Compared with control group,the level of interleukin (IL) 17E,IL 17F,interferon(IFN)γwere increased while the level of IL 4 was decreased in EAE group(P<0.05).Compared with the EAE group,the level of IFN γand IL 17E significantly reduced in Chaihu group.The difference has statistic significance(P<0.05 or P<0.01).Conclusion Chaihu has theapeutic effect on the EAE mice by anti inflammation.
