目的:观察片仔癀对局灶性脑梗死大鼠模型脑源性神经营养因子(BDNF)和白细胞介素6(IL 6)蛋白表达的影响。方法采用改良的 Longa法制备大脑中动脉栓塞(MCAO)模型,SD大鼠随机分为7组:假手术组、模型组、脑得生组、尼莫地平组及片仔癀大、中、小剂量组。通过免疫组化方法检测脑组织 BDNF和 IL 6的蛋白表达,HE染色观察脑组织的病理损伤情况。结果在梗死中心区,模型组的 BDNF和 IL 6蛋白表达显著低于假手术组(P<0.01),片仔癀各剂量组的 BDNF和 IL 6蛋白表达均高于模型组(P<0.05或P<0.01);在梗死周围区,模型组BDNF和IL 6蛋白表达均较假手术组显著升高,片仔癀各剂量组BDNF的蛋白表达均高于模型组(P<0.01),而 IL 6蛋白表达均低于模型组(P<0.05)。片仔癀大、中剂量组的脑组织病理形态较模型组有明显改善。结论片仔癀可以上调局灶性脑梗死后大鼠 BDNF的蛋白表达水平和下调 IL 6蛋白表达,减轻脑缺血导致的神经功能损伤。

목적:관찰편자황대국조성뇌경사대서모형뇌원성신경영양인자(BDNF)화백세포개소6(IL 6)단백표체적영향。방법채용개량적 Longa법제비대뇌중동맥전새(MCAO)모형,SD대서수궤분위7조:가수술조、모형조、뇌득생조、니막지평조급편자황대、중、소제량조。통과면역조화방법검측뇌조직 BDNF화 IL 6적단백표체,HE염색관찰뇌조직적병리손상정황。결과재경사중심구,모형조적 BDNF화 IL 6단백표체현저저우가수술조(P<0.01),편자황각제량조적 BDNF화 IL 6단백표체균고우모형조(P<0.05혹P<0.01);재경사주위구,모형조BDNF화IL 6단백표체균교가수술조현저승고,편자황각제량조BDNF적단백표체균고우모형조(P<0.01),이 IL 6단백표체균저우모형조(P<0.05)。편자황대、중제량조적뇌조직병리형태교모형조유명현개선。결론편자황가이상조국조성뇌경사후대서 BDNF적단백표체수평화하조 IL 6단백표체,감경뇌결혈도치적신경공능손상。
Objective To observe the effect of Pien Tze Huang(PTH)on expression of brain derived neurotrophic factor (BDNF) and interleukin 6(IL 6)protein in rats with focal cerebral infarction model.Methods The middle cerebral artery occlusion (MCAO) was established by the modified Longa method.Sprague Dawley(SD)rats were randomly divided into sham operation group,model group,high,medium and low dose PTH group,Naodesheng(NDS)group and nimodipine group.Pathological damage were observed by HE staining.Results In the central area of infarction,the expression of BDNF and IL 6 protein in model group were significantly lower than that in sham operation group.The expression of BDNF and IL 6 protein in Pien Tze Huang group were higher than those in the model group.In the area around the infarction,expression of BDNF and IL 6 protein in model group were significantly higher than those in the sham operation group,BDNF protein expression in PTH group were higher than those in the model group while the expression of IL 6 protein was lower than that in model group.Compared with model group,high and medium dose PTH groups had significant improvement on pathologial changes in brain tissue.Conclusion PTH can up regulated expression of BDNF protein and down regulation expression of IL 6 protein after focal cerebral infarction,al eviate nerve function injury caused by cerebral ischemia.
