目的:研究中药复方潜阳合剂对腹主动脉缩窄致高血压左室肥厚的影响,并探讨其作用机制。方法使用腹主动脉缩窄致高血压左室肥厚模型,Wistar大鼠随机分成假手术组和模型组,将高血压造模成功的大鼠随机分成模型组、培哚普利组和潜阳合剂组,每组10只,并开始灌胃,各组药物干预8周。使用心脏超声评价左室肥厚水平,尾动脉仪测量大鼠左前肢血压,天狼猩红染色观察心肌胶原含量,免疫组化法测定心肌组织Ⅰ型、Ⅲ型胶原纤维的含量。结果腹主动脉缩窄后12周,与假手术组相比,模型组血压升高,左室后壁及室间隔厚度增厚(P<0.001),左室舒张末内径和收缩末内径缩小(P<0.001);与模型组相比,培哚普利组和潜阳合剂组血压降低,左室后壁和室间隔厚度降低(P<0.001),舒张末内径(P<0.001)和收缩末内径(P<0.05)改善;与模型组相比,培哚普利组、潜阳合剂组大鼠心肌组织胶原较少,Ⅰ型和Ⅲ型胶原纤维表达减少。结论潜阳合剂可以改善腹主动脉缩窄致高血压大鼠的左室肥厚,其机制与降压、抑制心肌胶原纤维增生相关。

목적:연구중약복방잠양합제대복주동맥축착치고혈압좌실비후적영향,병탐토기작용궤제。방법사용복주동맥축착치고혈압좌실비후모형,Wistar대서수궤분성가수술조화모형조,장고혈압조모성공적대서수궤분성모형조、배타보리조화잠양합제조,매조10지,병개시관위,각조약물간예8주。사용심장초성평개좌실비후수평,미동맥의측량대서좌전지혈압,천랑성홍염색관찰심기효원함량,면역조화법측정심기조직Ⅰ형、Ⅲ형효원섬유적함량。결과복주동맥축착후12주,여가수술조상비,모형조혈압승고,좌실후벽급실간격후도증후(P<0.001),좌실서장말내경화수축말내경축소(P<0.001);여모형조상비,배타보리조화잠양합제조혈압강저,좌실후벽화실간격후도강저(P<0.001),서장말내경(P<0.001)화수축말내경(P<0.05)개선;여모형조상비,배타보리조、잠양합제조대서심기조직효원교소,Ⅰ형화Ⅲ형효원섬유표체감소。결론잠양합제가이개선복주동맥축착치고혈압대서적좌실비후,기궤제여강압、억제심기효원섬유증생상관。
Objective To study the effect and partial mechanism of Qiangyang decoction(QYD)on left ventricular hypertrophy(LVH) in rats with hypertension induced by abdominal aortic coarctation(AAC).Methods LVH model was established by AAC.Wistar rats were randomly divided into sham operation group and operation group.After hypertensive modeling,rats were randomly divided into model group,perindopril group and QYD group,there were ten rats in each group.After modeling,rats were gavaged for 8 weeks.LVH was determined by cardiac echocardiography,the blood pressure was determined by measuring left forelimb blood pressure with cau-dal artery continuous dynamic measurement instrument,myocardial col agen was observed by Sirius scarlet dye method,myocardial tissue type Ⅰ and type Ⅲ col agen fiber content was determined by immunohistochemistry.Results After 12 weeks of AAC,com-pared with the sham operation group,the systolic blood pressure,left ventricular posterior wal thickness (LVPWT)and interventric-ular septal thickness (IVST)were significantly increased in model group(P<0.001),while left ventricular end diastolic dimension (LVEDD)and left ventricular end systolic dimension (LVESD)were significantly decreased in(P<0.001).After intervention,com-pared with the model group,the systolic blood pressure,LVPWT and IVST of perindopril group and Qianyang decoction group are both significantly decreased(P<0.001),meanwhile,the LVEDD(P<0.001),and LVESD (P<0.05)were improved.Compared with the model group,myocardial tissue col agen fibrosis was less,type Ⅰ and type Ⅲ col agen expression was decreased.Conclusion QYD has effect against LVH in rats with hypertension induced by AAC,which is related with its antihypertension effect and inhibition of myocardial collagen fibrosis.
