目的：以荧光D-葡萄糖同系物（2-NBDG）为示踪探针，建立一种在活细胞中快速、直接检测糖摄入的敏感方法。方法通过培养肝癌细胞株HepG2，使用不同浓度的2-NBDG（0．3 mmol／L、1．0 mmol／L）处理20 min，用荧光显微镜观察细胞荧光强度，用IPP6．0软件分析细胞荧光强度。结果不同浓度2-NBDG（0．3 mmol／L、1．0 mmol／L）处理后，细胞荧光强度随剂量增加而显著增强（P＜0．05）。结论2-NBDG可以作为人源肝癌细胞株HepG2的荧光探测示踪剂进行糖摄入实验测定。本实验在细胞水平建立了简便可行的糖摄入能力评估方法，将有助于糖尿病的研究。
목적：이형광D-포도당동계물（2-NBDG）위시종탐침，건립일충재활세포중쾌속、직접검측당섭입적민감방법。방법통과배양간암세포주HepG2，사용불동농도적2-NBDG（0．3 mmol／L、1．0 mmol／L）처리20 min，용형광현미경관찰세포형광강도，용IPP6．0연건분석세포형광강도。결과불동농도2-NBDG（0．3 mmol／L、1．0 mmol／L）처리후，세포형광강도수제량증가이현저증강（P＜0．05）。결론2-NBDG가이작위인원간암세포주HepG2적형광탐측시종제진행당섭입실험측정。본실험재세포수평건립료간편가행적당섭입능력평고방법，장유조우당뇨병적연구。
Objetc vi e To develop a sensitive method for the rapid and direct detection of glucose uptake in human hepatocarcinoma HepG2 cells using a fluorescent D-glucose analog,2-NBDG,as a probe indicator.Methods HepG2 cell lines were cultured and then incubated by 2-NBDG of different concentrations(0.3 mmol/L1, .0mmol /L) for 20 minutes.The fluorescence intensity was observed via fluorescence imaging microscopy and analyzed using IPP6.0 software.Results The fluorescence intensity of incubated 2-NBDG of different concentration(s 0.3 mmol/L, 1.0 mmol/L) increased significantly with the increasing dosage of 2-NBDG(P<0.05).C onclusion 2-NBDG is a promising fluorescent probe indicator for the detection of glucose uptake in human hepatocarcinoma HepG2 cell lines.A simple and useful method for glucose uptake measurement at cell level is developed in the experiment,which might contribute to diabetes research.
