目的 探讨12周四氯化碳腹腔注射诱导小鼠肝纤维化的效果.方法 每周3次10%四氯化碳200μl腹腔注射,诱导C57BL/6小鼠肝脏纤维化.注射8、12周时处死小鼠,通过大体标本与石蜡切片镜检确定肝纤维化程度.结果 注射8周小鼠肝表面有颗粒感,镜下汇管区纤维向小叶内延伸,呈中度肝纤维化,注射12周小鼠肝表面出现结节,镜下有纤维隔伴假小叶形成,为重度肝纤维化.且12周四氯化碳腹腔注射对小鼠精神、体质量无明显影响,对照组体质量为29.0g,实验组为(26.8 ±0.7)g,差异无统计学意义(P>0.05),且实验中无小鼠死亡.结论 腹腔四氯化碳注射是一种安全有效的小鼠肝纤维化模型建立方法.

Objective To determine the hepatic fibrosis-induced effect of 12 week intraperitoneal injection of carbon tetrachloride in mice.Methods The hepatic fibrosis in C57BL/6 mice was induced by intraperitoneal injection of 200 μl 10% carbon tetrachloride three times every week.At 8th and 12th week,the mice were executed.The severity of hepatic fibrosis was identified by gross observation and microscopic examination.Results Livers of mice executed at week 8 had rough surface and fibers extended to the hepatic lobule,and moderate hepatic fibrosis was induced.In the mice executed at week 12,there were nodules on the liver surface,and the structure of hepatic lobules disordered and false lobules formed,suggesting the successful induction of hepatic fibrosis.Moreover,intraperitoneal injection of 12 week carbon tetrachloride had no significant influence on activity and mean weight of mice.Mean weight in control group and experimental group was 29.0 g and (26.8 ± 0.7) g respectively,P > 0.05.Conclusion Intraperitoneal injection of carbon tetrachloride in mice is a safe and effective method for building hepatic fibrosis model.
