选取海州常山( Clerodne drum trichotomum Thumb.)花粉为研究对象,研究了海州常山花粉活力测定的方法、最佳培养基配方及其最佳培养条件;探究了花期及花粉采集时间对海州常山花粉萌发的影响。结果表明:染色法及无机酸培养法不适宜于海州常山花粉活力的测定,最佳花粉活力测定法为液体萌发法;海州常山花粉萌发最佳培养基配方为150 g· L-1蔗糖+200 mg· L-1硼酸+150 mg·L -1氯化钙;其中硼酸及氯化钙pH值为5.5条件下花粉萌发效果更佳;花粉最适宜培养条件为干燥9,h 于25℃光照条件下液体培养8 h;花粉活力最强时间为花后1~2 d的08:00。

This experiment was conducted with C.trichotomum pollen to study the method of pollen activity assay , the best cul-ture medium formula and its optimal culture conditions and the effects of flowering and pollen collecting time on the pollen germination.The best methods of liquid staining and inorganic acid culture were not suitable for pollen activity assay , and the best method was liquid culture .The best culture medium formula for pollen germination was 150 g· L-1 sucrose+boric acid 200 mg· L-1+150 mg· L-1 calcium chloride .The effects of pollen germination would be better , if pH of boric acid and calcium chloride was5 .5.The appropriate culture conditions was as follows:drying pollen for 9 h, then putting it at 25℃with light liquid culture for 8 h.When C.trichotomumwas flowering for 1-2 d, the activity of pollen was the strongest .
