目的:观察丹参多酚酸盐联合聚维酮碘对大鼠术后肠粘连的影响,探讨其可能作用机制。方法选用SD雄性大鼠50只,随机分为对照组、模型组、丹参多酚酸盐组、聚维酮碘组(PVP-I组)及丹参多酚酸盐联合PVP-I组(联合组),每组10只。除对照组外,其余各组均制作肠粘连模型。对照组和模型组每天腹腔注射5%葡萄糖溶液;丹参多酚酸盐组、PVP-I组和联合组每天分别腹腔注射丹参多酚酸盐、PVP-I和二者联合,连续注射8天。大鼠于术后第7天开腹,肉眼观察各组大鼠术后粘连分级情况;酶联免疫吸附法(ELISA)检测各组大鼠血清和粘连组织白细胞介素-1β(IL-1β)、转化生长因子-β1(TGF-β1)水平。结果丹参多酚酸盐组及联合组肠粘连程度明显低于模型组(P<0.05);丹参多酚酸盐组、PVP-I组及联合组血清和粘连组织IL-1β表达水平均显著降低于模型组[(64.82±10.55)pg/mL、(70.93±16.06)pg/mL、(62.37±3.32)pg/mL vs(75.75±6.67)pg/mL,(178.90±17.28)pg/mL、(190.76±34.16)pg/mL、(168.40±25.14)pg/mL vs(220.78±18.31)pg/mL,P<0.05或P<0.01)。丹参多酚酸盐组、PVP-I组及联合组血清和粘连组织TGF-β1表达水平均显著降低于模型组[(36.77±5.25)pg/mL、(42.67±3.40)pg/mL、(21.90±1.15)pg/mL vs(54.93±4.75)pg/mL,(639.62±118.31)pg/mL、(645.32±56.95)pg/mL、(609.87±64.88)pg/mL vs(759.87±178.42)pg/mL,P<0.05或P<0.01)。结论丹参多酚酸盐、PVP-I及二者联合均能降低大鼠术后肠粘连程度,这可能与其降低血清和粘连组织中IL-1β和TGF-β1表达水平有关。

목적:관찰단삼다분산염연합취유동전대대서술후장점련적영향,탐토기가능작용궤제。방법선용SD웅성대서50지,수궤분위대조조、모형조、단삼다분산염조、취유동전조(PVP-I조)급단삼다분산염연합PVP-I조(연합조),매조10지。제대조조외,기여각조균제작장점련모형。대조조화모형조매천복강주사5%포도당용액;단삼다분산염조、PVP-I조화연합조매천분별복강주사단삼다분산염、PVP-I화이자연합,련속주사8천。대서우술후제7천개복,육안관찰각조대서술후점련분급정황;매련면역흡부법(ELISA)검측각조대서혈청화점련조직백세포개소-1β(IL-1β)、전화생장인자-β1(TGF-β1)수평。결과단삼다분산염조급연합조장점련정도명현저우모형조(P<0.05);단삼다분산염조、PVP-I조급연합조혈청화점련조직IL-1β표체수평균현저강저우모형조[(64.82±10.55)pg/mL、(70.93±16.06)pg/mL、(62.37±3.32)pg/mL vs(75.75±6.67)pg/mL,(178.90±17.28)pg/mL、(190.76±34.16)pg/mL、(168.40±25.14)pg/mL vs(220.78±18.31)pg/mL,P<0.05혹P<0.01)。단삼다분산염조、PVP-I조급연합조혈청화점련조직TGF-β1표체수평균현저강저우모형조[(36.77±5.25)pg/mL、(42.67±3.40)pg/mL、(21.90±1.15)pg/mL vs(54.93±4.75)pg/mL,(639.62±118.31)pg/mL、(645.32±56.95)pg/mL、(609.87±64.88)pg/mL vs(759.87±178.42)pg/mL,P<0.05혹P<0.01)。결론단삼다분산염、PVP-I급이자연합균능강저대서술후장점련정도,저가능여기강저혈청화점련조직중IL-1β화TGF-β1표체수평유관。
Objective To investigate the effects of salvianolate and povidone-iodine on postoperative intestinal adhesion in rats. Methods Fifty male SD rats were randomly divided into 5 groups: blank control group, model group, salvianolate group, povidone-iodine group and salvianolate combined with povidone-iodine group(combination group), with 10 rats in each group. A intestinal adhesion modeling was established in rats, and then each group were given the corresponding drugs(intraperitoneal injection of 5% glucose solution, 4mL/kg; salvianolate 10.9mg/kg;0.625% povidone-iodine, 4mL/kg) for 8d. On the 7th day, rats underwent open surgery in abdomen to observe and grade intestinal adhesion by macroscopy. The level of interleukin-1β and transform growth factor-β1 in blood sam-ples and adhesive tissue samples was determined by ELISA. Results Compared with model group, the grade of intestinal adhesion in salvianolate group and combination group was significantly lower (P<0.05);the level of IL-1βin blood samples and tissue samples in salvianolate group, povidone-iodine group and combination group were low-er(blood: 75.75±6.67pg/mL vs 64.82±10.55pg/mL, 70.93±16.06pg/mL, and 62.37±3.32pg/mL; tissue: 220.78±18.31pg/mL vs 178.90±17.28pg/mL, 190.76±34.16pg/mL, 168.40±25.14pg/mL; P<0.05 or P<0.01); the level of TGF-β1 in blood samples and tissue samples in salvianolate group, povidone-iodine group and combination group were lower (blood: 54.93±4.75pg/mL vs 36.77±5.25pg/mL, 42.67±3.40pg/mL, and 21.90±1.15pg/mL; tissue: 759.87±178.42pg/mL vs 639.62 ±118.31pg/mL, 645.32 ±56.95pg/mL, 609.87 ±64.88pg/mL; P<0.05 or P<0.01). Conclusion Both of sal-vianolate and povidone-iodine and their combination can effectively prevent intestinal adhesion after abdominal op-eration in rats, which may be related to reduction of IL-1β and TGF-β1 in peripheral blood and tissues.
