目的 制备门静脉高压大鼠模型,探讨血管生成素1(Ang-1)/酪氨酸激酶受体2(Tie-2)系统在门静脉高压大鼠食管壁组织的表达及其与食管壁新生血管形成的关系.方法 将50只雄性SD大鼠按体质量随机分为实验组和对照组,其中实验组30只(门静脉部分结扎+左肾上腺静脉结扎法制备门静脉高压模型),对照组20只(仅游离门静脉和左肾上腺静脉,不结扎),术后2周取大鼠食管下段组织标本,应用免疫组织化学技术检测食管壁Ang-1、Tie-2的表达情况,并计数CD34标记的微血管密度(MVD),分析Ang-1、Tie-2表达水平与食管壁血管新生的关系.结果 共37只大鼠门静脉高压模型建立成功(其中实验组17只,对照组20只).实验组Ang-1、Tie-2表达水平明显高于对照组,差异有统计学意义(P<0.05).其中Ang-1的表达水平与MVD无明显相关性(r =0.39,P>0.05),Tie-2的表达水平与MVD呈正相关(r=0.71,P<0.05).结论 门静脉高压大鼠食管壁组织中Ang-1、Tie-2、表达水平及MVD明显增高,Ang-1/Tie-2系统可能与门静脉高压大鼠模型食管壁组织的新生血管形成有关.

목적 제비문정맥고압대서모형,탐토혈관생성소1(Ang-1)/락안산격매수체2(Tie-2)계통재문정맥고압대서식관벽조직적표체급기여식관벽신생혈관형성적관계.방법 장50지웅성SD대서안체질량수궤분위실험조화대조조,기중실험조30지(문정맥부분결찰+좌신상선정맥결찰법제비문정맥고압모형),대조조20지(부유리문정맥화좌신상선정맥,불결찰),술후2주취대서식관하단조직표본,응용면역조직화학기술검측식관벽Ang-1、Tie-2적표체정황,병계수CD34표기적미혈관밀도(MVD),분석Ang-1、Tie-2표체수평여식관벽혈관신생적관계.결과 공37지대서문정맥고압모형건립성공(기중실험조17지,대조조20지).실험조Ang-1、Tie-2표체수평명현고우대조조,차이유통계학의의(P<0.05).기중Ang-1적표체수평여MVD무명현상관성(r =0.39,P>0.05),Tie-2적표체수평여MVD정정상관(r=0.71,P<0.05).결론 문정맥고압대서식관벽조직중Ang-1、Tie-2、표체수평급MVD명현증고,Ang-1/Tie-2계통가능여문정맥고압대서모형식관벽조직적신생혈관형성유관.
Objective To establish a rat model of portal hypertension and explore the relationship between the angiopoietin-1 (Ang-1)/tyrosine kinase receptors 2 (Tie-2) system and the neoangiogenesis in lower esophagus of rats with portal hypertension.Methods Fifty SD rats were randomly divided into two groups:experimental group (thirty rats,partial portal vein and left suprarenal vein were ligatured to make portal hypertension model) and control group (twenty rats,the portal vein and left suprarenal vein were separated but not ligatured).The lower esophageal tissue was obtained two weeks after the operation.The immunohistochemical streptavidin-perosidase (SP) technique was used to detect the expression of Ang-1,Tie-2 and CD34.The microvessel density (MVD) was determined by CD34 staining.The relationship between Ang-1,Tie-2 and the neoangiogenesis in lower esophagus was analyzed after that.Results The models of portal hypertension were set up successfully in 37 rats (17 in experimental group and 20 in control group).Compared with those of control group,the differences in the expression of Ang-1,Tie-2 and MVD were statistically significant (P < 0.05).The expression of Ang-1 had no significant correlation with MVD (r =0.39,P > 0.05),but the expression of Tie-2 was positively correlated with MVD (r=0.71,P<0.05).Conclusion The expression of Ang-1,Tie-2 and MVD is obviously higher in lower esophagus of rats in experimental group than that in the control group.The Ang-1/Tie-2 system is possibly related to the neoangiogenesis in lower esophagus of rats with portal hypertension.
