目的：通过构建针对窖蛋白（CAV）－1基因的siRNA，研究RNAi技术对MCF－7细胞cav－1基因表达的影响。方法构建siRNA表达载体pGenesil－1－shRNA－cav－1，通过脂质体法转染到MCF－7细胞，RT－PCR法检测cav－1的mRNA表达。结果成功构建重组表达载体pGenesil－1－shR－NA－cav－1；RT－PCR检测显示转染重组质粒的乳腺癌细胞中 cav－1 mRNA 表达明显低于阴性对照、空白对照和正常细胞组。结论重组表达载体pGenesil－1－shRNA－cav－1能有效抑制人乳腺癌细胞cav－1基因的表达。

목적：통과구건침대교단백（CAV）－1기인적siRNA，연구RNAi기술대MCF－7세포cav－1기인표체적영향。방법구건siRNA표체재체pGenesil－1－shRNA－cav－1，통과지질체법전염도MCF－7세포，RT－PCR법검측cav－1적mRNA표체。결과성공구건중조표체재체pGenesil－1－shR－NA－cav－1；RT－PCR검측현시전염중조질립적유선암세포중 cav－1 mRNA 표체명현저우음성대조、공백대조화정상세포조。결론중조표체재체pGenesil－1－shRNA－cav－1능유효억제인유선암세포cav－1기인적표체。
Objective To construct a new siRNA targeting to cav-1 applied to regulate the expression of cav-1 of MCF-7 cell lines. Methods The expression carrier pGenesil-1-shRNA-cav-1 was constructed,then the carrier was transfected into MCF-7 cell lines.The ex-pression of cav-1 mRNA was detected by reverse transcription PCR.Results It was successful to contruct the recombinated expression carri-er pGenesil-1-shRNA-cav-1.The expression of cav-1 mRNA in breast cells transfected recombination plasmids was obviously lower than the one in negative control,blank control and normal cells.Conclusions Recombinated carrier pGenesil-1-shRNA-cav-1 could effectively inhibit the expression of cav-1 in human breast cancer cell lines.
