目的：探讨4种常见阿尔茨海默病（AD）动物模型在研究凋亡机制领域中的优势。方法采用4种常见AD模型（东莨菪碱致记忆障碍模型、鹅膏蕈酸致痴呆大鼠模型、快速老化模型、双转基因小鼠模型），利用Morris水迷宫测试动物模型认知能力；免疫印迹检测Bcl－2蛋白表达水平；TUNEL染色检测凋亡水平。结果在凋亡机制研究探讨中，快速老化小鼠（SAM）在Morris水迷宫测试、Bcl－2蛋白表达以及凋亡检测中其正常组与模型组均有显著差异（P＜0．05）。结论 SAM模型在老年痴呆凋亡机制中有明显优势，为中药治疗老年痴呆的临床前研究提供指导。
목적：탐토4충상견아이자해묵병（AD）동물모형재연구조망궤제영역중적우세。방법채용4충상견AD모형（동랑탕감치기억장애모형、아고심산치치태대서모형、쾌속노화모형、쌍전기인소서모형），이용Morris수미궁측시동물모형인지능력；면역인적검측Bcl－2단백표체수평；TUNEL염색검측조망수평。결과재조망궤제연구탐토중，쾌속노화소서（SAM）재Morris수미궁측시、Bcl－2단백표체이급조망검측중기정상조여모형조균유현저차이（P＜0．05）。결론 SAM모형재노년치태조망궤제중유명현우세，위중약치료노년치태적림상전연구제공지도。
Objective To discuss advantages of 4 common AD animal models on research mechanism of apoptosis.Methods Four common AD model-scopolamine dysmnesia model,ibotenic acid dementia model,senescence accelerated model and double transgenic mice model were used.The cognitive competences of animal models were evaluated by Morris water maze.Bcl-12 protein expression was tested by Western blot and apoptosis was measured by immunohistochemistry TUNEL staining.Results The results of Morris water maze test,Bcl-12 protein expression and apoptosis had significant differences of SAM between normal and model groups(P<0.05).Conclusions SAM model has its great advantage in mechanism of apoptosis of AD ,so it could offer guidance in preclinical study of treating-AD herbs.
