建立高效液相色谱法分离测定“黑美人”马铃薯中原花青素含量的分析方法。实验采用C18柱(Phenomenex 5μm;250 mm ×4.6 mm);V(甲醇)∶V(0.5%冰醋酸水溶液)=30∶70为流动相;柱温:25℃;流速:1 mL/min;检测波长:287 nm。结果表明:原花青素在80μg/mL~200μg/mL浓度范围内有良好的线性关系,相关系数R=0.9957,方法的精密度RSD为2.86%,平均回收率为100.1%,原花青素的平均含量为51.805 mg/g。本方法简便、快速、准确、可靠,可用于“黑美人”马铃薯中原花青素的含量的测定。

Proanthocyanidins in “Heimeiren”Potatoes was determined by HPLC. Phenomenex C18 column (5μm;250 mm×4.6 mm) was used with the mobile phase of 30%methanol and 70 %water(with 0.5%acetic acid).The column temperature, flow rate and detection wavelength were 25 ℃, 1 mL/min and 287 nm, respectively. The results showed that there was a good linear relationship between peak area and proanthocyanidins content within the range of 80 μg/mL-200 μg/mL. Linear correlation was 0.995 7,RSD of precision was 2.86%,the average recovery rate was 100.1 %and the average content of proanthocyanidins was 51.805 mg/g. This method is simple, fast, accurate and reliable, suitable for determination of proanthocyanidins content in“Heimeiren”Potatoes.
