目的：研究大肠癌组织及癌旁正常组织 PTPRO 基因启动子甲基化状态及 mRNA 表达情况，并探讨其在大肠癌发生中的关系。方法：应用甲基化特异性 PCR（MSP）和 RT-PCR 分别检测30例大肠癌组织及癌旁正常组织中 PTPRO 基因启动子甲基化状态及 mRNA 表达状态。结果：大肠癌组织中 PTPRO 基因启动子甲基化率高于癌旁正常组织，其甲基化样本中 PTPRO 基因 mRNA 表达明显受抑。结论：PTPRO 基因启动子甲基化在大肠癌发生中起着重要作用，可能与大肠癌的发生、发展及预后有关。
目的：研究大腸癌組織及癌徬正常組織 PTPRO 基因啟動子甲基化狀態及 mRNA 錶達情況，併探討其在大腸癌髮生中的關繫。方法：應用甲基化特異性 PCR（MSP）和 RT-PCR 分彆檢測30例大腸癌組織及癌徬正常組織中 PTPRO 基因啟動子甲基化狀態及 mRNA 錶達狀態。結果：大腸癌組織中 PTPRO 基因啟動子甲基化率高于癌徬正常組織，其甲基化樣本中 PTPRO 基因 mRNA 錶達明顯受抑。結論：PTPRO 基因啟動子甲基化在大腸癌髮生中起著重要作用，可能與大腸癌的髮生、髮展及預後有關。
목적：연구대장암조직급암방정상조직 PTPRO 기인계동자갑기화상태급 mRNA 표체정황，병탐토기재대장암발생중적관계。방법：응용갑기화특이성 PCR（MSP）화 RT-PCR 분별검측30례대장암조직급암방정상조직중 PTPRO 기인계동자갑기화상태급 mRNA 표체상태。결과：대장암조직중 PTPRO 기인계동자갑기화솔고우암방정상조직，기갑기화양본중 PTPRO 기인 mRNA 표체명현수억。결론：PTPRO 기인계동자갑기화재대장암발생중기착중요작용，가능여대장암적발생、발전급예후유관。
Objective Research the PTPRO Gene promoter methylation and the expression of PTPRO gene mRNA in colorectal cancer tissue and normal tissue adjacent to carcinoma,and to discuss their relationship in colorectal cancer. Methods Methylation specific PCR (MSP) and RT-PCR were use to detected PTPRO gene promoter methylation status and mRNA’s expression in 30 cases of colorectal cancer tissue and in normal tissue adjacent to carcinoma. Results The PTPRO gene pro-moter methylation rate of colorectal cancer tissues is higher than normal tissue adjacent to carcinoma,the PTPRO gene mRNA’s expression of the methylation samples is obviously inhibitive. Conclusion The PTPRO gene promoter methylation plays an important role in colorectal cancer, and it may be related to the development and prognosis of colorectal cancer.
