目的:观察针刺干预对脑缺血大鼠脑组织缺氧诱导因子1α( hypoxia-inducible factor-1α,HIF-1α)蛋白及mRNA表达的影响,探讨针刺对缺血性脑血管疾病的防治机制。方法建立局灶性脑缺血大鼠模型,随机分为正常组、假手术组、模型组、针刺组(各8只大鼠),应用蛋白印迹及实时荧光定量检测针刺对局灶性脑缺血大鼠脑组织HIF-1α蛋白及mRNA表达的变化。结果正常组和假手术组大鼠比较HIF-1α蛋白及mRNA表达无明显变化,模型组大鼠HIF-1α蛋白及mRNA表达上调,差异无统计学意义;针刺组大鼠HIF-1α蛋白及mRNA表达高于模型组(0.567±0.058 vs 0.315±0.118;1.593±0.102 vs 1.193±0.259),差异有统计学意义(P<0.05)。结论脑缺血后大鼠脑组织HIF-1α蛋白及mRNA表达增强,针刺可以上调其表达从而发挥脑保护作用。
目的:觀察針刺榦預對腦缺血大鼠腦組織缺氧誘導因子1α( hypoxia-inducible factor-1α,HIF-1α)蛋白及mRNA錶達的影響,探討針刺對缺血性腦血管疾病的防治機製。方法建立跼竈性腦缺血大鼠模型,隨機分為正常組、假手術組、模型組、針刺組(各8隻大鼠),應用蛋白印跡及實時熒光定量檢測針刺對跼竈性腦缺血大鼠腦組織HIF-1α蛋白及mRNA錶達的變化。結果正常組和假手術組大鼠比較HIF-1α蛋白及mRNA錶達無明顯變化,模型組大鼠HIF-1α蛋白及mRNA錶達上調,差異無統計學意義;針刺組大鼠HIF-1α蛋白及mRNA錶達高于模型組(0.567±0.058 vs 0.315±0.118;1.593±0.102 vs 1.193±0.259),差異有統計學意義(P<0.05)。結論腦缺血後大鼠腦組織HIF-1α蛋白及mRNA錶達增彊,針刺可以上調其錶達從而髮揮腦保護作用。
목적:관찰침자간예대뇌결혈대서뇌조직결양유도인자1α( hypoxia-inducible factor-1α,HIF-1α)단백급mRNA표체적영향,탐토침자대결혈성뇌혈관질병적방치궤제。방법건립국조성뇌결혈대서모형,수궤분위정상조、가수술조、모형조、침자조(각8지대서),응용단백인적급실시형광정량검측침자대국조성뇌결혈대서뇌조직HIF-1α단백급mRNA표체적변화。결과정상조화가수술조대서비교HIF-1α단백급mRNA표체무명현변화,모형조대서HIF-1α단백급mRNA표체상조,차이무통계학의의;침자조대서HIF-1α단백급mRNA표체고우모형조(0.567±0.058 vs 0.315±0.118;1.593±0.102 vs 1.193±0.259),차이유통계학의의(P<0.05)。결론뇌결혈후대서뇌조직HIF-1α단백급mRNA표체증강,침자가이상조기표체종이발휘뇌보호작용。
Objective To observe the effect of acupuncture on hypoxia inducible factor -1αand local cerebral blood flow in rats with focal cerebral ischemia.Methods Wistar rats were randomly divided into four groups:normal group, sham-operated group, model group and electroacupuncture( EA) group.Focal cerebral ischemia rat models were developed by middle cerebral artery occlu-sion( MCAO ) with thread embolus.Electroacupuncture was applied to bilateral PC 6 and LI11 after MCAO.The expression of hypoxia inducible factor-1αwas examined in MCAO rats by Western blot and real-time quantitative PCR ( qPCR ) .Results Acupuncture could promote the recovery of neurological function in MCAO rats.Western blot and real-time qPCR showed that acupuncture promoted the hypoxia inducible factor-1αexpression in the ischemic brain tissue with statistical significance( P<0.05) .Conclusion Acupunc-ture promotes hypoxia inducible factor-1αexpression in rats with focal cerebral ischemia.All the results suggest that the increased ex-pression of hypoxia inducible factor-1αin the ischemic brain tissue plays an important role in brain protection.
