目的:研究缺氧诱导因子-1α在胆囊癌侵袭转移中的作用,并探讨 HIF-1α在判断胆囊癌治疗效果及预测胆囊癌复发转移方面的价值。方法:采用慢病毒对胆囊癌细胞系(GBC-SD)进行转染,干扰该细胞系中HIF-1α的表达,得到 HIF-1α表达不受干扰的胆囊癌细胞系(GBC-SD-NC)和 HIF-1α表达受干扰的胆囊癌细胞系 GBC-SD-HIF。通过将两组细胞系裸鼠鼠尾静脉注射建立胆囊癌血行转移模型,检测两组细胞在小鼠肺部形成转移瘤的能力。结果:GBC-SD-NC 组裸鼠肺部转移瘤数量、大小都比 GBC-SD-HIF 组高,GBC-SD-NC 组转移瘤的平均数量显著多于 GBC-SD-HIF 组。结论:干扰胆囊癌细胞中 HIF-1α的表达,可以显著减低胆囊癌细胞系GBC-SD 的血行肺转移能力。说明缺氧诱导因子-1α在胆囊癌侵袭转移中存在独立影响。
目的:研究缺氧誘導因子-1α在膽囊癌侵襲轉移中的作用,併探討 HIF-1α在判斷膽囊癌治療效果及預測膽囊癌複髮轉移方麵的價值。方法:採用慢病毒對膽囊癌細胞繫(GBC-SD)進行轉染,榦擾該細胞繫中HIF-1α的錶達,得到 HIF-1α錶達不受榦擾的膽囊癌細胞繫(GBC-SD-NC)和 HIF-1α錶達受榦擾的膽囊癌細胞繫 GBC-SD-HIF。通過將兩組細胞繫裸鼠鼠尾靜脈註射建立膽囊癌血行轉移模型,檢測兩組細胞在小鼠肺部形成轉移瘤的能力。結果:GBC-SD-NC 組裸鼠肺部轉移瘤數量、大小都比 GBC-SD-HIF 組高,GBC-SD-NC 組轉移瘤的平均數量顯著多于 GBC-SD-HIF 組。結論:榦擾膽囊癌細胞中 HIF-1α的錶達,可以顯著減低膽囊癌細胞繫GBC-SD 的血行肺轉移能力。說明缺氧誘導因子-1α在膽囊癌侵襲轉移中存在獨立影響。
목적:연구결양유도인자-1α재담낭암침습전이중적작용,병탐토 HIF-1α재판단담낭암치료효과급예측담낭암복발전이방면적개치。방법:채용만병독대담낭암세포계(GBC-SD)진행전염,간우해세포계중HIF-1α적표체,득도 HIF-1α표체불수간우적담낭암세포계(GBC-SD-NC)화 HIF-1α표체수간우적담낭암세포계 GBC-SD-HIF。통과장량조세포계라서서미정맥주사건립담낭암혈행전이모형,검측량조세포재소서폐부형성전이류적능력。결과:GBC-SD-NC 조라서폐부전이류수량、대소도비 GBC-SD-HIF 조고,GBC-SD-NC 조전이류적평균수량현저다우 GBC-SD-HIF 조。결론:간우담낭암세포중 HIF-1α적표체,가이현저감저담낭암세포계GBC-SD 적혈행폐전이능력。설명결양유도인자-1α재담낭암침습전이중존재독립영향。
Objective To study the effects of HIF-1αon the metastasis of gallbladder neoplasm, and discusses the value of HIF-1α on gallbladder neoplasm treatment. Methods Lentivirus infections were carried on to obtain the GBC-SD-NC whose expression of HIF-1α was normal,and GBC-SD-HIF that expression of HIF-1α was interfered. HIF-1α expression was in-terfered through vena caudalis in nude mice exenografts. HIF-1α expression effect was demonstrated on the metastasis ability of gallbladder neoplasm cells. Results GBC-SD-NC group whose number of metastasis and volume of metastasis were re-markably superior to the GBC-SD-HIF group. In In GBC-SD-NC group, the average number of metastasis foci permouse was marked more than GBC-SD-HIF group. Conclusion The interference of HIF-1α expression could lead to the decline of the metastasis ability of gallbladder neoplasm cells. The Results showed that HIF-1α could produce effects in on the metastasis of gallbladder neoplasm.
