目的::观察医用臭氧对神经病理性疼痛大鼠的镇痛作用并探讨其相关的中枢神经机制。方法:取成年雄性SD大鼠建立坐骨神经慢性压迫性损伤( CCI )模型;在损伤神经周围注射不同剂量医用臭氧后观察对 CCI 大鼠机械痛阈值(MWT)的影响;免疫组化技术检测CCI或医用臭氧对大鼠中脑导水管周围灰质外侧区(lPAG)中P2X3受体表达的影响。结果: CCI大鼠经医用臭氧治疗后MWT增高,疼痛缓解伴lPAG中P2X3受体表达上调;其MWT增高幅度和P2X3受体表达上调幅度与注射医用臭氧剂量成正相关;lPAG中微量注射P2X3受体特异性拮抗剂A-317491后部分翻转医用臭氧对CCI大鼠的镇痛作用。结论:损伤神经局部注射医用臭氧可对神经病理性疼痛大鼠产生镇痛作用,其镇痛机制与促进中枢lPAG中P2 X3受体的表达和功能活化有关。

목적::관찰의용취양대신경병이성동통대서적진통작용병탐토기상관적중추신경궤제。방법:취성년웅성SD대서건립좌골신경만성압박성손상( CCI )모형;재손상신경주위주사불동제량의용취양후관찰대 CCI 대서궤계통역치(MWT)적영향;면역조화기술검측CCI혹의용취양대대서중뇌도수관주위회질외측구(lPAG)중P2X3수체표체적영향。결과: CCI대서경의용취양치료후MWT증고,동통완해반lPAG중P2X3수체표체상조;기MWT증고폭도화P2X3수체표체상조폭도여주사의용취양제량성정상관;lPAG중미량주사P2X3수체특이성길항제A-317491후부분번전의용취양대CCI대서적진통작용。결론:손상신경국부주사의용취양가대신경병이성동통대서산생진통작용,기진통궤제여촉진중추lPAG중P2 X3수체적표체화공능활화유관。
Objective:To observe the analgesic effect of medical ozone on neuropathic pain in rats and a preliminary study of its mechanism. Methods:Using adult male Sprague Dawley ( SD) rats for the establishment of the rat sciatic nerve chronic constriction injury ( CCI) model,followed by injections of different doses of medical ozone around the injured sciatic nerve. Observing the changes of the mechanical withdrawal thresholds (MWT) and detected the expression alterations of P2X3 receptor in the rats’ lateral subdivision of midbrain lateral periaqueductal gray (lPAG). Results:The MWT values increased and the expressions of P2X3 receptor in the lPAG were up-regulated dose-dependently after CCI rats multiple injections of medical ozone. Intra-lPAG injection of P2X3 receptor antagonist A-317491 partially but significantly reversed the analgesic effect of medical ozone. Conclusion:The injured-nerve local injections of medical ozone can improve the mechanical pain threshold values of neuropathic pain rats which is associated with the activation of P2X3 receptors in the lPAG.
