目的:观察奥美拉唑对反流性食管黏膜损伤后大鼠血浆和食管组织中细胞因子 IL-6及TNF-α的影响,探讨奥美拉唑治疗反流性食管炎的作用机制。方法:选用健康雄性SD大鼠,体重220~250g,随机将动物分为5组,假手术组、单纯胃食管反流动物模型组(模型组)、模型组+奥美拉唑低剂量组、中剂量组、高剂量组。分别取食管标本肉眼观察黏膜状况。应用ELISA方法检测大鼠血浆和食管组织中细胞因子IL-6及TNF-α的变化。结果:假手术组大鼠食管颜色淡红,有光泽,黏膜光滑。模型组大鼠食管颜色变淡白,黏膜粗糙、增厚、皱襞灰白色斑块状增生。与模型组比较,奥美拉唑低、中、高剂量治疗组大鼠食管病变明显减轻。 ELISA检测结果显示,假手术组大鼠血浆和食管组织有一定IL-6及TNF-α的基础含量。模型组大鼠血浆和食管组织IL-6及TNF-α含量明显升高,奥美拉唑治疗组大鼠血浆和食管组织IL-6及TNF-α含量明显降低。结论:奥美拉唑明显降低反流性食管黏膜损伤后大鼠血浆和食管组织炎症因子IL-6及TNF-α的含量,可能是其改善食管局部及外周炎症反应、治疗反流性食管炎的重要作用机制。

목적:관찰오미랍서대반류성식관점막손상후대서혈장화식관조직중세포인자 IL-6급TNF-α적영향,탐토오미랍서치료반류성식관염적작용궤제。방법:선용건강웅성SD대서,체중220~250g,수궤장동물분위5조,가수술조、단순위식관반류동물모형조(모형조)、모형조+오미랍서저제량조、중제량조、고제량조。분별취식관표본육안관찰점막상황。응용ELISA방법검측대서혈장화식관조직중세포인자IL-6급TNF-α적변화。결과:가수술조대서식관안색담홍,유광택,점막광활。모형조대서식관안색변담백,점막조조、증후、추벽회백색반괴상증생。여모형조비교,오미랍서저、중、고제량치료조대서식관병변명현감경。 ELISA검측결과현시,가수술조대서혈장화식관조직유일정IL-6급TNF-α적기출함량。모형조대서혈장화식관조직IL-6급TNF-α함량명현승고,오미랍서치료조대서혈장화식관조직IL-6급TNF-α함량명현강저。결론:오미랍서명현강저반류성식관점막손상후대서혈장화식관조직염증인자IL-6급TNF-α적함량,가능시기개선식관국부급외주염증반응、치료반류성식관염적중요작용궤제。
Objective:To observe the effect of omeprazole on the level of IL-6 and TNF-αin the rats model of esophageal mucosal injury induced by reflux of acid .Method:Healthy male SD rats weighing 220 to 250g were randomly divided into 5 groups: sham-operated group(sham), gastroesophageal reflux group (model), omeprazole low dose, middle dose and high dose groups .Rat models of reflux esophagitis were in-duced by single acid reflux .The contents of IL-6 and TNF-αwere detected by ELISA staining .Result:The esophagus of rat was erythroid and lustrous , with glossy mucosal membranes in sham group .Compared with that of the sham group , the esophagus presented light white and the esophageal mucosal membrane became rough, thicken, grey plaque-like proliferation in rats of model group .Compared with that of model group , a-bove the changes were significantly decreased in omeprazole treatment groups .Compared with that of thesham group , the contents of IL-6 and TNF-αwere significantly increased in model group .Compared with thats of the model group , the contents of IL-6 and TNF-αwere significantly decreased in treatment of ome-prazole group .Conclusion:It could be concluded that omeprazole could decrease the esophgeal mucosal in -jury induced by reflux of acid by decrease the contents of IL-6 and TNF-α.
