目的:观察新生大鼠坏死性小肠结肠炎肠道组织含半胱氨酸的天冬氨酸蛋白水解酶3(Caspase3)与增殖细胞核抗原(PCNA)表达,探讨新生大鼠坏死性小肠结肠炎(NEC)疾病过程中Caspase3和PCNA表达的意义及相关性。方法新生SD大鼠24只随机分为空白对照组和NEC模型组,每组12只。空白对照组予鼠乳代乳品人工喂养3 d;NEC模型组采用代乳品喂养+缺氧+冷刺激,连续3 d建立NEC模型。3d后处死,采用免疫组织化学方法检测各组大鼠肠组织Caspase3和PCNA的表达,两组间比较釆用Wilcoxon秩和检验,并对NEC模型组肠组织Caspase3和PCNA的表达行Spearman相关检验。结果 NEC模型组肠组织Caspase3表达指数(77.3±8.65)%明显高于空白对照组(18.92±3.37)%,差异有统计学意义(P=0.0000)。而NEC模型组肠组织PCNA表达指数(15.00±1.94)%明显低于空白对照组(34.17±5.78)%、差异有统计学意义(P=0.0001),NEC模型组肠道组织病理评分与Caspase3的表达指数(P=0.005)、Caspase3/PCNA表达指数比(P=0.0016)呈正相关;而与PCNA表达指数呈负相关(P=0.014)。结论 Caspase3可能通过表达上调参与了NEC的疾病过程,而PCNA可能通过下调表达量参与了NEC的疾病过程,细胞增殖和凋亡失衡对NEC的发生发展有一定关系。

목적:관찰신생대서배사성소장결장염장도조직함반광안산적천동안산단백수해매3(Caspase3)여증식세포핵항원(PCNA)표체,탐토신생대서배사성소장결장염(NEC)질병과정중Caspase3화PCNA표체적의의급상관성。방법신생SD대서24지수궤분위공백대조조화NEC모형조,매조12지。공백대조조여서유대유품인공위양3 d;NEC모형조채용대유품위양+결양+랭자격,련속3 d건립NEC모형。3d후처사,채용면역조직화학방법검측각조대서장조직Caspase3화PCNA적표체,량조간비교변용Wilcoxon질화검험,병대NEC모형조장조직Caspase3화PCNA적표체행Spearman상관검험。결과 NEC모형조장조직Caspase3표체지수(77.3±8.65)%명현고우공백대조조(18.92±3.37)%,차이유통계학의의(P=0.0000)。이NEC모형조장조직PCNA표체지수(15.00±1.94)%명현저우공백대조조(34.17±5.78)%、차이유통계학의의(P=0.0001),NEC모형조장도조직병리평분여Caspase3적표체지수(P=0.005)、Caspase3/PCNA표체지수비(P=0.0016)정정상관;이여PCNA표체지수정부상관(P=0.014)。결론 Caspase3가능통과표체상조삼여료NEC적질병과정,이PCNA가능통과하조표체량삼여료NEC적질병과정,세포증식화조망실형대NEC적발생발전유일정관계。
Objective To study the expressions of Cysteinyl aspartate specific proteinase 3(Caspase3) and proliferating cell nuclear antigen(PCNA)in intestinal tissue of neonate rats with necrotizing enterocolitis,in order to explore their significance and their correlation. Methods Randomly selected 24 neonate rats at the age of 48 hours,were divided into 2 groups.12 rats every group,Blank control group and NEC model group. Blank control group rats were given artificial feeding with self-control milk. NEC model group rats were given artificial feeding with self-control milk and cold exposure after hypoxic-reoxygen twice a day for 3 days.Detected the expression of Caspase3 and PCNA by the method of immunohistochemical . Results The Caspase3 expression index of intestinal tissue in NEC model group (77.3±8.65)%was significantly higher than the control group (18.92±3.37)%,and the difference was statistically significant (P=0.0000). While the PCNA expression index of the intestinal tissue in the NEC model group(15±1.94)%was significantly lower than the control group (34.17±5.78)%,and there was statistical significance (P=0.0001).The correlation were positive Between the Caspase3 expression(P=0.005),Caspase3/PCNA ratio (P=0.0016)and pathological score of the intestinal tissues in the NEC model group.They showed negative correlation between the PCNA expression ,and pathological score of the intestinal tissues in the NEC model group (P=0.5321).Conclusions The Caspase3 maybe involved in the NEC’s disease process by up regulating its expression and PCNA maybe involved in the disease process of NEC by down-regulating its expression. The balance between apoptosis and proliferation should be destroyed during the disease process of NEC.
