目的:观察金思维提取物(GAPT)对 APPswe/PS1dE9双转基因小鼠海马内 APP 表达水平的影响。方法将3月龄的 APPswe/PS1dE9转基因小鼠随机分为模型组、盐酸多奈哌齐组及 GAPT 大、中、小剂量组,并以同月龄遗传背景相同的 C57BL/6J 小鼠作为正常组,每组12只,灌胃给药6个月后,以蛋白印迹法(Western blot)检测海马内淀粉样前体蛋白(APP)表达变化。结果模型组小鼠海马中 APP 表达水平显著高于正常组(P <0.01);各治疗组与模型组相比,APP 蛋白表达水平均显著降低,其中 GAPT 小剂量组(P <0.01)、 GAPT 中剂量组(P <0.01)、GAPT 大剂量组(P <0.01)、多奈哌齐组(P <0.01);各 GAPT 组与多奈哌齐组比较结果 APP 表达水平均高于多奈哌齐组,其中 GAPT小剂量组(P <0.01)、 GAPT 中剂量组(P <0.01)有显著性差异,而 GAPT 大剂量组(P >0.05)无明显差异。结论GAPT 可能具有抑制 APPswe/PS1dE9双转基因小鼠海马内 APP 蛋白过度表达的作用。
目的:觀察金思維提取物(GAPT)對 APPswe/PS1dE9雙轉基因小鼠海馬內 APP 錶達水平的影響。方法將3月齡的 APPswe/PS1dE9轉基因小鼠隨機分為模型組、鹽痠多奈哌齊組及 GAPT 大、中、小劑量組,併以同月齡遺傳揹景相同的 C57BL/6J 小鼠作為正常組,每組12隻,灌胃給藥6箇月後,以蛋白印跡法(Western blot)檢測海馬內澱粉樣前體蛋白(APP)錶達變化。結果模型組小鼠海馬中 APP 錶達水平顯著高于正常組(P <0.01);各治療組與模型組相比,APP 蛋白錶達水平均顯著降低,其中 GAPT 小劑量組(P <0.01)、 GAPT 中劑量組(P <0.01)、GAPT 大劑量組(P <0.01)、多奈哌齊組(P <0.01);各 GAPT 組與多奈哌齊組比較結果 APP 錶達水平均高于多奈哌齊組,其中 GAPT小劑量組(P <0.01)、 GAPT 中劑量組(P <0.01)有顯著性差異,而 GAPT 大劑量組(P >0.05)無明顯差異。結論GAPT 可能具有抑製 APPswe/PS1dE9雙轉基因小鼠海馬內 APP 蛋白過度錶達的作用。
목적:관찰금사유제취물(GAPT)대 APPswe/PS1dE9쌍전기인소서해마내 APP 표체수평적영향。방법장3월령적 APPswe/PS1dE9전기인소서수궤분위모형조、염산다내고제조급 GAPT 대、중、소제량조,병이동월령유전배경상동적 C57BL/6J 소서작위정상조,매조12지,관위급약6개월후,이단백인적법(Western blot)검측해마내정분양전체단백(APP)표체변화。결과모형조소서해마중 APP 표체수평현저고우정상조(P <0.01);각치료조여모형조상비,APP 단백표체수평균현저강저,기중 GAPT 소제량조(P <0.01)、 GAPT 중제량조(P <0.01)、GAPT 대제량조(P <0.01)、다내고제조(P <0.01);각 GAPT 조여다내고제조비교결과 APP 표체수평균고우다내고제조,기중 GAPT소제량조(P <0.01)、 GAPT 중제량조(P <0.01)유현저성차이,이 GAPT 대제량조(P >0.05)무명현차이。결론GAPT 가능구유억제 APPswe/PS1dE9쌍전기인소서해마내 APP 단백과도표체적작용。
Objective To investigate the effect of GAPT, a combination of herbal extracts , on amyloid precursor protein (APP) in AP-Pswe /PS1dE9 double transgenic mice.Methods Sixty APPswe/PS1dE9 transgenic mice aged 3 month after birth were randomly divided in-to three GAPT groups by intragastric administration at doses of 1.22 g/kg, 2.44 g/kg, 4.88 g/kg per day, and a donepezil group by intra-gastric administration at doses of 0.92 mg/kg per day, an APPswe/PS1dE9 transgenic model group and other twelve C 57BL/6J mice aged 3 month after birth as normal control by intragastric administration of sodium carboxy -methylcellulose (CMC) solution.A six -month treat-ment regimen with GAPT was administered to APP /PS1 transgenic mice.The changes of expressions of APP were detected by using Western blot method.Results The detection showed that the expressions of APP increased significantly in model group compared with normal group . Compared with model group, the expressions of APP in three GAPT groups and donepezil group decreased significantly , and in low -dose GAPT group (P <0.01), in mid -dose GAPT group (P <0.01), in high -dose GAPT group (P <0.01), in donepezil group (P <0.01).Compared with normal group, the expressions of APP in treatment groups increased , in low -dose GAPT group (P <0.01) and in mid -dose GAPT group (P <0.01), among them there was significant difference ; However in high -dose GAPT group (P >0.05) and in donepezil group (P >0.05), there was no significant difference ; The result of three different dose groups of GAPT showed significantly dif -ferent in low -dose GAPT group (P <0.01) and in mid -dose GAPT group (P <0.01) from donepezil group.Conclusion This study sμggested that GAPT could play a role of inhibiting the over -expression of APP in hippocampi area of APP /PS1 transgenic mice.
