目的:观察HT‐29细胞在低氧下分泌的基质金属蛋白酶2/9(MMP‐2/9)活性,检测NF‐κB的蛋白表达,初步探讨低氧与肿瘤恶性演进的关系。方法参照体内肿瘤的氧分压,建立低氧模型。将 H T‐29细胞低氧处理不同时段,明胶酶谱分析检测其分泌的MMP‐2/9活性变化;提取细胞核蛋白,Western blot测定胞核内NF‐κB的表达。结果低氧12 h ,HT‐29细胞分泌的MMP‐2/9活性逐渐上调,24 h达顶峰。在低氧的诱导下,HT‐29细胞核内NF‐κB的表达趋势与MMP‐2/9活性变化趋势基本一致。结论低氧能诱导 HT‐29细胞分泌高活性的 MMP‐2/9而促进其向恶性演进,可能与细胞核内 NF‐κB的表达上调有关。
目的:觀察HT‐29細胞在低氧下分泌的基質金屬蛋白酶2/9(MMP‐2/9)活性,檢測NF‐κB的蛋白錶達,初步探討低氧與腫瘤噁性縯進的關繫。方法參照體內腫瘤的氧分壓,建立低氧模型。將 H T‐29細胞低氧處理不同時段,明膠酶譜分析檢測其分泌的MMP‐2/9活性變化;提取細胞覈蛋白,Western blot測定胞覈內NF‐κB的錶達。結果低氧12 h ,HT‐29細胞分泌的MMP‐2/9活性逐漸上調,24 h達頂峰。在低氧的誘導下,HT‐29細胞覈內NF‐κB的錶達趨勢與MMP‐2/9活性變化趨勢基本一緻。結論低氧能誘導 HT‐29細胞分泌高活性的 MMP‐2/9而促進其嚮噁性縯進,可能與細胞覈內 NF‐κB的錶達上調有關。
목적:관찰HT‐29세포재저양하분비적기질금속단백매2/9(MMP‐2/9)활성,검측NF‐κB적단백표체,초보탐토저양여종류악성연진적관계。방법삼조체내종류적양분압,건립저양모형。장 H T‐29세포저양처리불동시단,명효매보분석검측기분비적MMP‐2/9활성변화;제취세포핵단백,Western blot측정포핵내NF‐κB적표체。결과저양12 h ,HT‐29세포분비적MMP‐2/9활성축점상조,24 h체정봉。재저양적유도하,HT‐29세포핵내NF‐κB적표체추세여MMP‐2/9활성변화추세기본일치。결론저양능유도 HT‐29세포분비고활성적 MMP‐2/9이촉진기향악성연진,가능여세포핵내 NF‐κB적표체상조유관。
Objective To investigate the correlation between moderate hypoxia and tumor invasiveness in colon carcinoma cell line HT‐29 and detect the expression of NF‐κB protein in nucleus and activities of MMP‐2/9 ,so as to explore its latent mecha‐nism of malignant transformation .Methods The moderate hypoxia model was established according to the pressure of O2 of tumor in vivo .After different period in moderate hypoxia ,activities of MMP‐2/9 secreted by HT‐29 cells were measured by gelatin zymog‐raphy ,NF‐κB levels in nucleus were assessed by Western blot after extraction of nucleic protein .Results MMP‐2/9 activities under moderate hypoxia gradually up‐regulated in 12 h and reached the peak at 24 h .The expression of NF‐κB protein in nucleus was simi‐lar to that of MMP‐2/9 .Conclusion Moderate hypoxia could induce malignant phenotype in HT‐29 cells through up‐regulating MMP‐2/9 activities ,in which NF‐κB may involve in this progression .
