目的:探讨吡格列酮对高糖培养乳鼠心肌细胞炎症的抑制作用。方法 SD乳鼠原代心肌细胞培养至90%融合,换上无血清的DMEM培养基,并随机将细胞分为正常组、高糖模型组、吡格列酮低剂量组(10μmol/L)和高剂量组(20μmol/L),继续培养48h后,分别收集细胞培养液及心肌细胞,检测培养液中乳酸脱氢酶(LDH)、炎症因子IL-6、TNF-α的含量以及心肌细胞中蛋白P38MAPK的表达量和NF-κB的活性。结果高糖下的细胞经吡格列酮药物处理后LDH、IL-6、TNF-α的含量、P38MAPK的表达量和NF-κB的活性均明显下降。结论吡格列酮能抑制高糖培养下心肌细胞炎症作用。

목적:탐토필격렬동대고당배양유서심기세포염증적억제작용。방법 SD유서원대심기세포배양지90%융합,환상무혈청적DMEM배양기,병수궤장세포분위정상조、고당모형조、필격렬동저제량조(10μmol/L)화고제량조(20μmol/L),계속배양48h후,분별수집세포배양액급심기세포,검측배양액중유산탈경매(LDH)、염증인자IL-6、TNF-α적함량이급심기세포중단백P38MAPK적표체량화NF-κB적활성。결과고당하적세포경필격렬동약물처리후LDH、IL-6、TNF-α적함량、P38MAPK적표체량화NF-κB적활성균명현하강。결론필격렬동능억제고당배양하심기세포염증작용。
Objective To investigate the effect of pioglitazone on high glucose-induced inflammation in neonatal rat cardiac myocytes .Methods Ventricular myocytes were isolated from neonatal rats and cultured in DMEM medium.All the cells were randomly divided into 3 groups:control group,high glucose group,tpioglita-zone low-dose group and pioglitazone high-dose group .The levels of LDH ,IL-6 and TNF-αin cardiac myocytes were determined.At the same time,the expression of p38MAPK and NF-κB protein was examined by Western blot .Results High glucose induced obvious injury of cardiac myocytes ,increased the contents of LDH ,IL-6 and TNF-αand elevated the expression of p 38MAPK and NF-κB protein.Treatment with pioglitazone could signifi-cantly inhibite high glucose-induced inflammatory injury ,decrease the contents of LDH ,IL-6 and TNF-α,and reversed the elevation of p 38 MAPK and NF-κB protein expression induced by high glucose .Conclusion Piogli-tazone can inhibit the inflammatory reactionof cultured neonatal rat cardiac myocytes induced by high glucose .
