用酸提取法从鲫鱼中提取胶原蛋白,用不同浓度的盐酸作为脱钙溶液,以0.5 mol/L乙酸—乙酸钠缓冲溶液提取其胶原蛋白。单因素分析与正交试验结果表明:提取温度为6~10℃时,最佳提取条件为固液比1砄20,提取时间每次1.5 h,脱钙酸浓度为0.5 mol/L,脱钙时间3 h,胶原蛋白提取率为0.773%。
用痠提取法從鯽魚中提取膠原蛋白,用不同濃度的鹽痠作為脫鈣溶液,以0.5 mol/L乙痠—乙痠鈉緩遲溶液提取其膠原蛋白。單因素分析與正交試驗結果錶明:提取溫度為6~10℃時,最佳提取條件為固液比1砄20,提取時間每次1.5 h,脫鈣痠濃度為0.5 mol/L,脫鈣時間3 h,膠原蛋白提取率為0.773%。
용산제취법종즉어중제취효원단백,용불동농도적염산작위탈개용액,이0.5 mol/L을산—을산납완충용액제취기효원단백。단인소분석여정교시험결과표명:제취온도위6~10℃시,최가제취조건위고액비1결20,제취시간매차1.5 h,탈개산농도위0.5 mol/L,탈개시간3 h,효원단백제취솔위0.773%。
Using hydrochloric acid with different concentrations as decalcification solution and the mixture of acetic and sodium acetate as buffer solution , the collagen wan extracted from erucian fish scale.The univariate analysis and orthogonal test results showed that the extraction ratio was 0.773% under the optimal conditions: the ratio of solid to solvent as 1:20, the extracting time as 1.5 days, the concentration of hydrochloric acid for decalcification as 0.5 mol/L and the decalcification times as 3 h.
