Paclitaxel%neuropathic pain%animal model%rats
目的:建立紫杉醇致大鼠神经病理性疼痛模型。方法40只成年雄性 SD 大鼠随机分为4组,每组10例,每天测定大鼠的机械缩足反射阈值(MWT),隔天腹腔注射紫杉醇,剂量分别为0、0.5、1.0、2.0 mg/(kg·d),共10次,第20 d 显微镜观察神经结构。结果腹腔注射紫杉醇2.0 mg/kg 组大鼠最早在第3次注射后(第6 d)出现机械痛敏,并在第4次注射后(第8 d)机械痛阈降到最低。腹腔注射紫杉醇2.0 mg/kg 组大鼠的坐骨神经髓鞘肿胀,部分髓鞘空泡变,部分许旺细胞结构破坏,许旺细胞核增多。结论间断重复腹腔注射2.0 mg/(kg·d)紫杉醇可以成功建立 SD 大鼠外周神经病理性疼痛模型。

목적:건립자삼순치대서신경병이성동통모형。방법40지성년웅성 SD 대서수궤분위4조,매조10례,매천측정대서적궤계축족반사역치(MWT),격천복강주사자삼순,제량분별위0、0.5、1.0、2.0 mg/(kg·d),공10차,제20 d 현미경관찰신경결구。결과복강주사자삼순2.0 mg/kg 조대서최조재제3차주사후(제6 d)출현궤계통민,병재제4차주사후(제8 d)궤계통역강도최저。복강주사자삼순2.0 mg/kg 조대서적좌골신경수초종창,부분수초공포변,부분허왕세포결구파배,허왕세포핵증다。결론간단중복복강주사2.0 mg/(kg·d)자삼순가이성공건립 SD 대서외주신경병이성동통모형。
Objective To establish a rat model of neuropathic pain induced by Paclitaxel. Methods For-ty Sprague-Dawley adult male rats were randomly divided into 4 groups (n =10 each group).Paclitaxel [0, 0.5,1.0,2.0 mg/(kg·d)]or vehicle was administered intraperitoneally every 2 days respectively until 10 injec-tions had been given.Mechanical withdrawal threshold (MWT)of rats'hind paw were measured everyday.The microstructures of sciatic nerve were observed by using microscope on the 20th day after injection. Results Mechanical hyperalgesia of rats first appeared after the 3rd injection (on day 6 )with 2.0 mg/ kg Paclitaxel, then decreased to minimum after the 4th injection (on day 8).Rats of intraperitoneal injection with 2.0 mg/kg Paclitaxel group had sciatic nerve myelin swelling,parts of myelin showed vacuolar degeneration,parts of Schwann cell structures were destroyed,the number of Schwann cells was increased. Conclusion A SD rat peripheral neuropathy pain model induced by paclitaxel can be successively established by intraperitoneally in-jecting 2.0 mg/kg Paclitaxel with intermittent repetition.
