目的:探究盐酸吗啉胍片(ABOB)对心肌细胞的毒性。方法将小鼠随机分为盐酸吗啉胍片高剂量组(8只)、盐酸吗啉胍片低剂量组(8只)、生理盐水对照组(8只),给小鼠灌胃给药7 d后,检测小鼠血清中肌酸激酶(CK)、脑钠肽(BNP)、肌钙蛋白 T (CTnT)的水平;HE染色对心脏、肾脏的病理学形态分析。同时对新生大鼠心肌细胞培养,用MTT法检测盐酸吗啉胍片对心肌细胞的抑制作用。结果与对照组比较,盐酸吗啉胍片高剂量组、盐酸吗啉胍片低剂量组的C K水平增高,差异有统计学意义(P<0.05),BNP和CTnT水平差异无统计学意义;与对照组比较,盐酸吗啉胍片高剂量组、盐酸吗啉胍片低剂量组的心肌组织切片呈病理样改变,肾脏组织未见明显变化;与空白对照组比较,给药组心肌细胞活性受到不同程度的抑制,并随着给药剂量的增加,心肌细胞活性逐渐下降。结论盐酸吗啉胍片对心肌细胞有一定的毒性。

목적:탐구염산마람고편(ABOB)대심기세포적독성。방법장소서수궤분위염산마람고편고제량조(8지)、염산마람고편저제량조(8지)、생리염수대조조(8지),급소서관위급약7 d후,검측소서혈청중기산격매(CK)、뇌납태(BNP)、기개단백 T (CTnT)적수평;HE염색대심장、신장적병이학형태분석。동시대신생대서심기세포배양,용MTT법검측염산마람고편대심기세포적억제작용。결과여대조조비교,염산마람고편고제량조、염산마람고편저제량조적C K수평증고,차이유통계학의의(P<0.05),BNP화CTnT수평차이무통계학의의;여대조조비교,염산마람고편고제량조、염산마람고편저제량조적심기조직절편정병리양개변,신장조직미견명현변화;여공백대조조비교,급약조심기세포활성수도불동정도적억제,병수착급약제량적증가,심기세포활성축점하강。결론염산마람고편대심기세포유일정적독성。
Objective To explore the myocardial cell toxicity of Moroxydine Hydrochloride Tablets(ABOB) .Methods Mice were randomly divided into ABOB high dose group (8) ,low dose group (8) ,and saline control group (8) .Mice serum creatine kinase (CK) ,brain natriuretic peptide (BNP) ,CTnT levels were determined 7 d after intragastric administration .The hearts and kidneys were removed and processed for histological examination .At the same time ,the inhibition effect of ABOB on the newborn rat my‐ocardial cell was evaluated by MTT assay .Results Compared with the control group ,ABOB high dose group ,ABOB low dose group significantly increased the levels of CK (P<0 .05) ,while no obvious change was observed in the levels of BNP and CTnT . Compared with the control group ,ABOB high dose group and ABOB low dose group ,myocardial tissue presented patholoical change ,while no obvious change found in kidney tissue .Compared with the control group ,myocardial cell activity was inhibited in different degrees after administration of ABOB with high or low dose ,and tended to decline gradually with increasing the drug dose .Conclusion ABOB has certain toxicity for myocardial cells .
