将乳鼠心肌细胞分为对照组、高糖组、波动性高糖组以及波动性高糖+NF‐κB特异性抑制剂吡咯烷二硫代氨基甲盐酸(PDTC)组,培养6天后进行心肌细胞的鉴定、心肌细胞活性、心肌细胞凋亡、NF‐κB和p53的蛋白表达的检测。结果显示,与高糖组相比,波动性高糖组的细胞存活率下降,凋亡率、NF‐κB和p53的表达增多。与波动性高糖+PDTC组相比,波动性高糖组细胞存活率下降,凋亡率、NF‐κB和p53的表达增多。因此在波动性高糖条件下,NF‐κB和p53表达更易增多,NF‐κB可能通过p53介导心肌细胞凋亡。

장유서심기세포분위대조조、고당조、파동성고당조이급파동성고당+NF‐κB특이성억제제필각완이류대안기갑염산(PDTC)조,배양6천후진행심기세포적감정、심기세포활성、심기세포조망、NF‐κB화p53적단백표체적검측。결과현시,여고당조상비,파동성고당조적세포존활솔하강,조망솔、NF‐κB화p53적표체증다。여파동성고당+PDTC조상비,파동성고당조세포존활솔하강,조망솔、NF‐κB화p53적표체증다。인차재파동성고당조건하,NF‐κB화p53표체경역증다,NF‐κB가능통과p53개도심기세포조망。
Cardiac muscle cells from SD rats were cultured and randomly divided into control group ,high glucose group , intermittent high glucose group and intermittent high glucos + PDTC group .After 6 days ,the myocardial cells were identified viability , apoptosis of cardiac muscle cells were detected ,as well as expression levels of NF‐κB and p53 . Compared with the high glucose group ,in cardiac muscle cells which were exposed in intermittent high glucose group ,the viability was significantly lower ,the apoptosis rate and the levels of NF‐κB and p53 were obviously higher .The viability of cardiac muscle cells was higher ,the apoptosis rate and the levels of NF‐κB and p53 of cardiac muscle cells were lower in intermittent high glucose+ PDTC group than in intermittent high glucose group .So the expression levels of NF‐κB and p53 in intermittent high glucose conditions are higher than in continued high glucose conditions .
