【目的】研究转 AtGAPC2基因马铃薯试管苗对低磷胁迫的响应,评价并筛选耐低磷优良株系。【方法】以26个转 AtGAPC2基因马铃薯株系及其非转基因对照品种“川芋10号”的试管苗为材料,在组培体系下设置正常磷(1.25×10-3 mol/L)与低磷(1.25×10-5 mol/L)两个水平,测定相关指标。【结果】结果表明:低磷胁迫下,相比于“川芋10号”,转 AtGAPC2基因株系株型正常,部分表现出地上部生长势旺、根系发达、试管薯产量高等特征,转基因株系间在形态学方面亦存在差异;运用模糊隶属函数法将所有株系划分为4个级别,96.15%转 AtGAPC2基因株系的隶属函数平均值高于“川芋10号”,其中以第一级别中 L13、L1、L3、L18、L19这5个株系表现最好;合单株块茎产量的综合值,最终筛选出 L13、L1、L3这3个株系,其单株块茎产量综合值分别是56.2%、93.0%、83.1%,显著高于“川芋10号”的26.1%。【结论】转 AtGAPC2基因能提高马铃薯试管苗对低磷胁迫的耐受性。
【目的】研究轉 AtGAPC2基因馬鈴藷試管苗對低燐脅迫的響應,評價併篩選耐低燐優良株繫。【方法】以26箇轉 AtGAPC2基因馬鈴藷株繫及其非轉基因對照品種“川芋10號”的試管苗為材料,在組培體繫下設置正常燐(1.25×10-3 mol/L)與低燐(1.25×10-5 mol/L)兩箇水平,測定相關指標。【結果】結果錶明:低燐脅迫下,相比于“川芋10號”,轉 AtGAPC2基因株繫株型正常,部分錶現齣地上部生長勢旺、根繫髮達、試管藷產量高等特徵,轉基因株繫間在形態學方麵亦存在差異;運用模糊隸屬函數法將所有株繫劃分為4箇級彆,96.15%轉 AtGAPC2基因株繫的隸屬函數平均值高于“川芋10號”,其中以第一級彆中 L13、L1、L3、L18、L19這5箇株繫錶現最好;閤單株塊莖產量的綜閤值,最終篩選齣 L13、L1、L3這3箇株繫,其單株塊莖產量綜閤值分彆是56.2%、93.0%、83.1%,顯著高于“川芋10號”的26.1%。【結論】轉 AtGAPC2基因能提高馬鈴藷試管苗對低燐脅迫的耐受性。
【목적】연구전 AtGAPC2기인마령서시관묘대저린협박적향응,평개병사선내저린우량주계。【방법】이26개전 AtGAPC2기인마령서주계급기비전기인대조품충“천우10호”적시관묘위재료,재조배체계하설치정상린(1.25×10-3 mol/L)여저린(1.25×10-5 mol/L)량개수평,측정상관지표。【결과】결과표명:저린협박하,상비우“천우10호”,전 AtGAPC2기인주계주형정상,부분표현출지상부생장세왕、근계발체、시관서산량고등특정,전기인주계간재형태학방면역존재차이;운용모호대속함수법장소유주계화분위4개급별,96.15%전 AtGAPC2기인주계적대속함수평균치고우“천우10호”,기중이제일급별중 L13、L1、L3、L18、L19저5개주계표현최호;합단주괴경산량적종합치,최종사선출 L13、L1、L3저3개주계,기단주괴경산량종합치분별시56.2%、93.0%、83.1%,현저고우“천우10호”적26.1%。【결론】전 AtGAPC2기인능제고마령서시관묘대저린협박적내수성。
Abstrzct:[Objective]The aim of the study was to study the responses of the test-tube plantlets of transgenic AtGAPC2 potato to low phosphorus stress and to screen the excellent lines with resist-ance to low phosphorus.[Method]26 transgenic lines and no-transgenic "Chuanyu 10" varieties were cultured in two phosphorus levels (1.25 mmol/L,0.0125 mmol/L).In addition,the rele-vant indicators were determined.[Results]Compared to "Chuanyu 10",transgenic lines showed faster growth,more developed root system and higher production under low phosphorus stress. Those lines were divided into four levels by fuzzy subordination method.96.15% transgenic lines performed better than"Chuanyu 10"did.The first level including five strains (L13,L1,L3,L18 and L19)was best.Combination with the comprehensive value of tuber yield per plant,L13,L1 and L3 three strains were eventually selected.The comprehensive value of tuber yield per plant were 56.2%,93.0% and 83.1% respectively,which were 26.1% higher than that of "Chuanyu 10".[Conclusion]The AtGAPC2 gene can improve the tolerance of test-tube plantlets to low phosphorus stress.
