运用正交试验和单因素比较试验,探讨半夏蛋白提取过程中各条件对蛋白质量浓度的影响,优化半夏总蛋白的分离纯化工艺.用DEAE‐Sepharose FF离子交换层析对半夏凝集素进行分离、纯化、凝血鉴定,并进行抑菌效果的探究.建立半夏总蛋白的最佳提取工艺:称取适量新鲜半夏块茎,超声波处理20 min ,用1.5% NaCl溶液在4℃下静置抽提6h,提取1次后离心取上清并进行硫酸铵沉淀得到半夏总蛋白.凝集素对金黄色葡萄球菌的抑制效果明显,对大肠杆菌的抑制效果一般.

The result of univariate test and orthogonal comparative test haveshowed the influence of various conditions on pinellia total protein's concentration during the extraction process .According to the result , the better extraction way of Pinellia total protein has been conducted in this study :firstly ,weighed some fresh Pinellia tubers ,treat them with ultrasonic wave for 20 min ,extracted one time with 1 .5% NaCl so‐lution for 6h at 4 ℃ ,then took the supernatant after centrifuging ,finally Pinellia total protein was got af‐ter ammonium sulphate precipitation .The lectin was extracted from the Pinellia total protein through DE‐AE‐Sepharose FF ion‐exchange liquid chromatography ,then the lectin's hemagglutination activity and its antibacterial effect was detected .This antibacterial test shows that the inhibitory effect of Pinellia lectin on Staphylococcus aureusis significant ,but its inhibitory effect on Escherichia coli is general .
