为给制定临床使用抗菌药物的轮换用药方案,降低耐药性的危害提供科学依据,采用单因素试验和正交试验方法,研究不同细菌传代、营养因素和耐药菌下大肠杆菌耐药菌株 D600对环丙沙星、恩诺沙星耐药水平的影响。结果表明:D600经210代传代,对环丙沙星的最小抑菌浓度(MIC)由128μg/mL(高度耐药)降至32μg/mL(中度耐药),而对恩诺沙星的 MIC 值没有变化;在稀释32倍的 LB 肉汤培养基中传代,D600对环丙沙星、恩诺沙星的 MIC 值由≥128μg/mL(高度耐药)分别降至0.25μg/mL 和1μg/mL(敏感)。营养因素水平越低、传代次数越多、耐药菌比例越小,D600对药物的 MIC 值越小,直至恢复到敏感。且上述因子对环丙沙星敏感性恢复的作用强于恩诺沙星。

To provide a scientific basis for reducing the harm of drug resistance and formulating clinical rotational schedule of antibiotics application,whether subculturing,nutrition factors and resistant bacteria strain D600 would raise E.coli sensitivity to ciprofloxacin and enrofloxacin were investigated by using the methods of single factor test and orthogonal test.The results indicated when continued subcultivation to 210 generations,the MIC value of E.coli D600 to ciprofloxacin dropped from 128 μg/mL (high-leve resistance)to 32 μg/mL(mind-leve resistance),but its MIC value to enrofloxacin not changed.The MIC values of E.coli D600 to two drugs,which was subcultrued 210 times in lower nutrition level of broth, dropped from high-leve resistance (128 μg/mL)to sensitivity (ciprofloxacin 0.25 μg/mL,enrofloxacin 0.5 μg/mL).The lower nutrition level of culture medium,the more subcultivation times and lesser proportions of resistance strains,the lower the MIC values of E.coli to the antibiotics were up to be sensitivity.The three factors tested showed certain effect on the drug-sensitivity restoration of E.coli to ciprofloxacin and enrofloxacin,furthermore the effect to ciprofloxacin was stronger than to enrofloxacin.
