为建立液体培养真菌菌丝体及发酵液的高效分离方法,以不同种属具代表性的真菌菌株(木霉属的棘孢木霉菌、毛壳属的球毛壳菌、核盘菌属的核盘菌、镰刀菌属的尖孢镰刀菌、链格孢属的链格孢菌和丝核菌属的立枯丝核菌)为例,采用高速离心法、减压抽滤法和多层纱布过滤法分别将菌丝体与发酵液分离,比较3种分离方法的分离效果。结果表明,高速离心和减压抽滤法操作步骤多、收集速度慢、耗电、成本高、对某些液体培养菌丝体收集效果差;多层纱布过滤法操作简单、分离速度快,节能、成本低以及收集的菌丝体含水量低,且不影响提取菌丝体的核酸质量。

In order to set up a high effective method for separation of fungal mycelium and its fermentation broth, six kinds of typical fungi, which from different species including Trichoderma asperellum ,Chaetomium globosum ,Sclerotinia sclerotiorum,Fusarium oxysporum ,Alternalia alternate and Rhizoctonia solani ,were selected for the experiments.Three methods of high-speed centrifugation, vacuum filteration and multi-layer gauze filtration were employed to separate fungal mycelium and its fermentation broth.Results:The methods of the high-speed centrifugation and vacuum filtration has some problems,such as operating complexity,collecting slowly,power consumption,high cost and inferior effectiveness for collected fungal mycelium of some species.However,the multilayer gauze filter method has many advantages including easy operation,separating fast,energy conservation,low cost and low moisture content of harvested mycelium.In addition,the mycelium quality settles for extracting nucleonic acid.
