目的:通过观测大鼠异位骨化模型中低氧诱导因子-1α( hypoxia inducible factor-1α, HIF-1α)及血管内皮生长因子(vascular endothelial growth factor, VEGF)表达变化,探讨HIF-1α、VEGF信号通路在异位骨化发生发展中的作用。方法 SD大鼠60只随机分为实验组和对照组,各30只,实验组制备异位骨化模型。于5、10周取跟腱组织, RT-PCR及免疫印迹试验(Westen-blot)检测两组局部组织中HIF-1α及VEGF表达水平。结果术后5、10周对照组HIF-1α及VEGF表达量均显著低于实验组(P<0.05)。结论在大鼠跟腱异位骨化早期, HIF-1α及VEGF表达明显升高,提示其对异位骨化发生、发展有重要作用。

목적:통과관측대서이위골화모형중저양유도인자-1α( hypoxia inducible factor-1α, HIF-1α)급혈관내피생장인자(vascular endothelial growth factor, VEGF)표체변화,탐토HIF-1α、VEGF신호통로재이위골화발생발전중적작용。방법 SD대서60지수궤분위실험조화대조조,각30지,실험조제비이위골화모형。우5、10주취근건조직, RT-PCR급면역인적시험(Westen-blot)검측량조국부조직중HIF-1α급VEGF표체수평。결과술후5、10주대조조HIF-1α급VEGF표체량균현저저우실험조(P<0.05)。결론재대서근건이위골화조기, HIF-1α급VEGF표체명현승고,제시기대이위골화발생、발전유중요작용。
Objective To observe expression changes of hypoxia inducible factor-1α(HIF-1α) and vascular endothelial growth factor (VEGF) in rat Achilles tendon heterotopic ossification model, and to investigate effects of HIF-1αand VEGF signal path in occurrence and progression of heterotopic ossification. Methods A total of 60 SD rats were randomly divided into experimental group and control group, and each group contained 30 cases. Heterotopic ossification model was made in the experimental group. Achilles tendon tissues were taken in 5 and 10 weeks for detection of HIF-1αand VEGF expression levels by RT-PCR and Western-blot test. Results In 5 and 10 weeks after operation, the control group had obviously lower HIF-1αand VEGF expression levels than the experimental group (P<0.05). Conclusion Remarkably increased HIF-1αand VEGF expression levels in early stage of rat Achilles tendon heterotopic ossification provides important effects for occurrence and progression of heterotopic ossification.
