目的 探讨赖氨酸特异性去甲基酶1 (Lysine specific demethylase 1,LSD1)的表达水平对结肠癌LoVo细胞黏附能力的影响及其分子机制.方法 选取人结肠癌LoVo细胞株,通过LSD1-shRNA质粒瞬时转染人结肠癌LoVo细胞株,RT-PCR和Western blot检测处理后LoVo细胞中KSD1的表达情况,黏附试验检测转染后LoVo细胞的同质性黏附和异质性黏附能力变化,RT-PCR检测处理后LoVo细胞TGF-β1、HES-1、VEGF-C、CD44v6、MMP7、β-catenin等分子的mRNA水平变化.多个样本比较采用单因素方差分析,两两比较采用t检验.结果 LSD1-shRNA质粒瞬时转染人结肠癌LoVo细胞株1h及2h后,细胞的同质性黏附能力均增加(F=151.52和96.47,均P<0.05),异质性黏附能力下降(F=87.54和81.50,均P<0.05);RT-PCR检测显示VEGF-C、CD44v6、MMP7和β-catenin基因表达水平下降(均P<0.05).结论 LSD1下调可以引起LoVo细胞黏附能力改变,过程涉及到β-catenin分子信号通路.

목적 탐토뢰안산특이성거갑기매1 (Lysine specific demethylase 1,LSD1)적표체수평대결장암LoVo세포점부능력적영향급기분자궤제.방법 선취인결장암LoVo세포주,통과LSD1-shRNA질립순시전염인결장암LoVo세포주,RT-PCR화Western blot검측처리후LoVo세포중KSD1적표체정황,점부시험검측전염후LoVo세포적동질성점부화이질성점부능력변화,RT-PCR검측처리후LoVo세포TGF-β1、HES-1、VEGF-C、CD44v6、MMP7、β-catenin등분자적mRNA수평변화.다개양본비교채용단인소방차분석,량량비교채용t검험.결과 LSD1-shRNA질립순시전염인결장암LoVo세포주1h급2h후,세포적동질성점부능력균증가(F=151.52화96.47,균P<0.05),이질성점부능력하강(F=87.54화81.50,균P<0.05);RT-PCR검측현시VEGF-C、CD44v6、MMP7화β-catenin기인표체수평하강(균P<0.05).결론 LSD1하조가이인기LoVo세포점부능력개변,과정섭급도β-catenin분자신호통로.
Objective To investigate the effect of lysine specific demethylase 1 (LSD1) expression on adhesion ability of LoVo cell and related molecular mechnism.Methods After colon cancer LoVo cell lines being transfected LSD1-shRNA plasmids,RT-PCR and Western blot were used to detect the expression of LSD1;Adhesion test to detect the homogeneity adhesion and heterogeneous adhesion abilities.RT-PCR to detect the mRNA level change of TGF-beta1,hypothesis-1,VEGF-C,CCR7,MMP7 and beta-catenin in LoVo cells.Results LSD1-shRNA plasmids were transfected into LoVo cells after 1 h and 2 h respectively,cell heterogeneous adhesion ability declined (F =151.52,96.47,P < 0.05).Homogeneity adhesion ability enhanced (F =87.54,81.50,P < 0.05).The gene expression of VEGF-C,CD44v6,MMP7 and β-catenin decreased obviously (P < 0.05).Conclusions Downregulted LSD1 expression inhibits colon cancer LoVo cell heterogeneous adhesion abilities and promotes homogeneity adhesion abilities.The mechanism may be related to beta-catenin molecular signal pathway.
