Atp5a1%comparative proteomics%Esophageal Cancer
目的:利用比较蛋白质组学方法获得差异表达蛋白,从中寻找与新疆哈萨克族食管癌发生、发展密切相关的基因,为新疆哈萨克族食管癌的早期诊断和治疗提供依据。方法利用二维凝胶电泳(2-dimensionelectro-phoresis,2-DE)和基质辅助激光解吸电离飞行时间质谱(matrix-assisted laser desorption/ionization time-of-flight mass spectrometry,MALDI-TOF-MS)技术联用检测新疆哈萨克族食管癌癌组织与远端正常组织的差异表达蛋白,对其中表达上调的 Atp5a1基因进行 RT-PCR 验证。结果2-DE 结果显示新疆哈萨克族食管癌癌组织中Atp5a1明显可见,蛋白表达强度差异>1.5倍。RT-PCR 验证结果显示,癌组织中 Atp5a1 mRNA 明显升高,差异有统计学意义(P <0.05)。结论Atp5a1基因的高表达和食管癌的发生、发展密切相关。

목적:이용비교단백질조학방법획득차이표체단백,종중심조여신강합살극족식관암발생、발전밀절상관적기인,위신강합살극족식관암적조기진단화치료제공의거。방법이용이유응효전영(2-dimensionelectro-phoresis,2-DE)화기질보조격광해흡전리비행시간질보(matrix-assisted laser desorption/ionization time-of-flight mass spectrometry,MALDI-TOF-MS)기술련용검측신강합살극족식관암암조직여원단정상조직적차이표체단백,대기중표체상조적 Atp5a1기인진행 RT-PCR 험증。결과2-DE 결과현시신강합살극족식관암암조직중Atp5a1명현가견,단백표체강도차이>1.5배。RT-PCR 험증결과현시,암조직중 Atp5a1 mRNA 명현승고,차이유통계학의의(P <0.05)。결론Atp5a1기인적고표체화식관암적발생、발전밀절상관。
Objective To obtain differential expression proteins of Kazakhs esophageal cancer in Xinjiang by a comparative proteomics method to find closely related gene;to provide clues for early diagnosis and ther-apy of occurrence and development of Kazakhs esophageal cancer Xinjiang.Methods Two-dimensional gel electrophoresis (2-DE)and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS)were utilized for the identification of proteins differentially expressed between cancer and adjacent non-cancerous tissue.The differential expression of Atp5a1 was further verified by RT-PCR. Results 2-DE results showed that kazakhs esophageal carcinoma tissue Atp5a1 clearly visible and the in-tensity of protein expression differences greater than 1.5 times in Xinjiang.RT-PCR validation results showed that cancer tissue Atp5a1 mRNA increased obviously,the difference was statistically significant (P <0.05).Conclusion Over expression of Atp5a1 gene might be a significant function of occurrence and development of Kazakhs esophageal cancer in Xinjiang.
