为建立‘中秋酥脆枣’的组培快繁体系,对外植体消毒时间长短和种类选择、不定芽诱导、不定芽增殖、不定根诱导进行研究。结果表明:最佳外植体是水培枝枣头;该外植体用升汞处理5 min后,接种于MS(改良)+6-BA 2.5 mg/L+NAA 0.15 mg/L,不定芽诱导率达94%;最适增殖培养基为MS(改良)+6-BA 1.5 mg/L+NAA 0.5 mg/L,平均不定芽达6.5;1/2MS(改良)+NAA 0.15 mg/L为最适生根培养基,不定芽平均生根3.6个。

위건립‘중추소취조’적조배쾌번체계,대외식체소독시간장단화충류선택、불정아유도、불정아증식、불정근유도진행연구。결과표명:최가외식체시수배지조두;해외식체용승홍처리5 min후,접충우MS(개량)+6-BA 2.5 mg/L+NAA 0.15 mg/L,불정아유도솔체94%;최괄증식배양기위MS(개량)+6-BA 1.5 mg/L+NAA 0.5 mg/L,평균불정아체6.5;1/2MS(개량)+NAA 0.15 mg/L위최괄생근배양기,불정아평균생근3.6개。
In order to establish the propagation system of‘Zhongqiusucui zao’, the sterilizing time of explants, kind choosing of explants, induction and proliferation of adventitious buds and roots were studied. The result showed that the bud explants from water-cultured branches was the best explant. This kind of explant could be sterilized by 0.1%HgCl2 for 5 min;MS (ameliorated) medium with 2.5 mg/L 6-BA and 0.15 mg/L NAA was the optimum starting medium. The induction rate of sprout was 94%. MS (ameliorated) medium with 1.5 mg/L 6-BA and 0.5 mg/L NAA was the optimum proliferation medium. The sprout number per explant amounted to 6.5. The optimum rooting medium was 1/2 MS (ameliorated) with 0.15 mg/L NAA. Sprout had an average root number of 3.6.
