目的:研究超支化聚缩水甘油-羟喜树碱药物递送系统(biodegradable hyperbranched polyglycerol-hydroxy camp-tothecin, dHPG-HA)对体外培养的人腺样囊性癌(高肺转移)细胞(salivary adenoid cystic carcinoma-lung metastases, SACC-LM)的抑制作用。方法:细胞计数试剂盒(cell counting kit, CCK8)检测药物浓度为0.001、0.01、0.1、1、10、100μg/mL的羟喜树碱(hydroxy camptothecin, HA)和含等量HA的dHPG-HA,在24、48 h对肿瘤细胞的抑制作用;流式细胞仪检测浓度为0.001、0.01、0.1、1、10μg/mL的dHPG-HA是否能诱导SACC-LM细胞凋亡。结果:HA和dHPG-HA对SACC-LM细胞增殖均有明显抑制作用,且作用呈剂量依赖性和时间依赖性;dHPG-HA对SACC-LM细胞的细胞毒性明显低于HA;dHPG-HA能诱导SACC-LM细胞凋亡。结论:dHPG的构建能达到使药物HA缓释的作用并降低其细胞毒性。

목적:연구초지화취축수감유-간희수감약물체송계통(biodegradable hyperbranched polyglycerol-hydroxy camp-tothecin, dHPG-HA)대체외배양적인선양낭성암(고폐전이)세포(salivary adenoid cystic carcinoma-lung metastases, SACC-LM)적억제작용。방법:세포계수시제합(cell counting kit, CCK8)검측약물농도위0.001、0.01、0.1、1、10、100μg/mL적간희수감(hydroxy camptothecin, HA)화함등량HA적dHPG-HA,재24、48 h대종류세포적억제작용;류식세포의검측농도위0.001、0.01、0.1、1、10μg/mL적dHPG-HA시부능유도SACC-LM세포조망。결과:HA화dHPG-HA대SACC-LM세포증식균유명현억제작용,차작용정제량의뢰성화시간의뢰성;dHPG-HA대SACC-LM세포적세포독성명현저우HA;dHPG-HA능유도SACC-LM세포조망。결론:dHPG적구건능체도사약물HA완석적작용병강저기세포독성。
Objective: To investigate the inhibition effects of hyperbranched polyglycidol-hydroxy camptothecin (dHPG-HA) drug delivery system on cultured human adenoid cystic carcinoma (high lung metastasis) cells (SACC-LM). Methods:SACC-LM cells cultured in vitro were treated with different concentrations of hydroxy camptothecin (HA) and (dHPG-HA) which contained equal amounts of HA, and the proliferation inhibition of SACC-LM cells was detected with cell counting kit assay (CCK8). Apoptosis of SACC-LM cells treated by various concentrations of HA and dHPG-HA was ascertained by flow cytometry. Results:The proliferation of SACC-LM cells was significantly reduced. And the effect was dose-dependent and time-dependent. In addition, the cytotoxicity of dHPG-HA on SACC-LM cells was obviously lower than free HA. Fur-thermore, dHPG-HA could induce apoptosis of SACC-LM cells effectively. In a word, the construction of drug carrier (dH-PG) on HA provided sustained release of drugs and reduced its cytotoxicity.
