目的:建立反相高效液相色谱法测定大鼠体内表柔比星血药浓度的方法。方法色谱柱为 Agilent tc - C18;流动相为:0,01 moL/ L 磷酸二氢铵缓冲溶液(冰醋酸调 pH =4,4):乙腈=79:21;检测波长:254 nm;流速:1,0 ml/ min;进样量:20μl;柱温:室温。选择柔红霉素作为内标,以蛋白质沉淀法提取血浆样品。结果检测表柔比星血药浓度在0,01172~10,0μg/ ml 范围内线性关系良好(r =0,9995),最低定量限为0,01172μg/ ml(S/ N≥10);日内 RSD <5,75%,日间 RSD <4,8%;样品平均回收率为96,3%,RSD 为4.8%(n =5);表柔比星血浆样品在72 h 内不同条件下基本稳定。结论反相高效液相色谱法测定大鼠体内表柔比星的血药浓度准确可靠,灵敏度高、专属性好,可用于表柔比星血药浓度测定。

Objective To establish the method for the determination of in vivo epirubicin deter-mined with reversed phase high performance liquid chromatography(RH - HPLC)in rats, Methods Chrom-atographic column:Agilent tc - C18 ,mobile phase:the ratio of 0, 01 mol/ L ammonium dihydrogen phosphate buffer solution(mixed with glacial acetic acid pH = 4, 4)and acetonitrile was 79: 21;detection wavelength:254 nm;flow rate:1, 0 ml/ min;sample size:20 μl;column temperature:room temperature, Daunorubicin was selected as internal standard, The protein precipitation method was used for the extraction of plasma sample, Results In the range of 0, 01172 to 10, 0μg/ ml,the plasma concentration of epirubicin presented the good linear relationship(r = 0, 999 5),and the minimum quantitative limit was 0, 011 72 μg/ ml(S/ N≥10), The intraday RSD was < 5, 75% and the daytime RSD was < 4, 8% , The average recovery of sample was 96, 3% ,RSD = 4, 8%(n = 5), In 72 h,the plasma sample of epirubicin was basically stable under different conditions, Conclusion RH - HPLC is accurate,reliable,highly sensitive and well specific in the determi-nation of plasma concentration of epirubicin in the rats in vivo, This method is applicable for the determina-tion of plasma concentration of epirubicin.
