目的:考察黄芩提取物( SBE)对D-半乳糖和亚硝酸钠合并诱导的阿尔茨海默病(AD)模型小鼠海马及皮质乙酰胆碱酯酶(AChE)和胆碱乙酰转移酶( ChAT)的影响。方法小鼠腹腔注射D-半乳糖150 mg? kg-1和亚硝酸钠100 mg? kg-1制备AD模型,随机将小鼠分为5组,正常组与模型组和3个剂量实验组,实验组分别灌胃给予SBE(125,250,500 mg? kg-1? d-1),连续60 d。在给药第60天,将小鼠断头取脑,测定皮质和海马中的蛋白含量,用试剂盒测定小鼠海马和皮质中AChE、ChAT的活性。结果3个剂量SEB可剂量依赖性的升高AD模型小鼠海马和皮质中ChAT的活性,并降低AChE的活性。中、高2个剂量实验组与模型组相比,可不同程度增强海马中AChE、ChAT的活力。结论 SBE可促进AD模型小鼠海马和皮质中乙酰胆碱的合成并抑制其分解,提示SBE具有改善AD小鼠学习与记忆的功能。
目的:攷察黃芩提取物( SBE)對D-半乳糖和亞硝痠鈉閤併誘導的阿爾茨海默病(AD)模型小鼠海馬及皮質乙酰膽堿酯酶(AChE)和膽堿乙酰轉移酶( ChAT)的影響。方法小鼠腹腔註射D-半乳糖150 mg? kg-1和亞硝痠鈉100 mg? kg-1製備AD模型,隨機將小鼠分為5組,正常組與模型組和3箇劑量實驗組,實驗組分彆灌胃給予SBE(125,250,500 mg? kg-1? d-1),連續60 d。在給藥第60天,將小鼠斷頭取腦,測定皮質和海馬中的蛋白含量,用試劑盒測定小鼠海馬和皮質中AChE、ChAT的活性。結果3箇劑量SEB可劑量依賴性的升高AD模型小鼠海馬和皮質中ChAT的活性,併降低AChE的活性。中、高2箇劑量實驗組與模型組相比,可不同程度增彊海馬中AChE、ChAT的活力。結論 SBE可促進AD模型小鼠海馬和皮質中乙酰膽堿的閤成併抑製其分解,提示SBE具有改善AD小鼠學習與記憶的功能。
목적:고찰황금제취물( SBE)대D-반유당화아초산납합병유도적아이자해묵병(AD)모형소서해마급피질을선담감지매(AChE)화담감을선전이매( ChAT)적영향。방법소서복강주사D-반유당150 mg? kg-1화아초산납100 mg? kg-1제비AD모형,수궤장소서분위5조,정상조여모형조화3개제량실험조,실험조분별관위급여SBE(125,250,500 mg? kg-1? d-1),련속60 d。재급약제60천,장소서단두취뇌,측정피질화해마중적단백함량,용시제합측정소서해마화피질중AChE、ChAT적활성。결과3개제량SEB가제량의뢰성적승고AD모형소서해마화피질중ChAT적활성,병강저AChE적활성。중、고2개제량실험조여모형조상비,가불동정도증강해마중AChE、ChAT적활력。결론 SBE가촉진AD모형소서해마화피질중을선담감적합성병억제기분해,제시SBE구유개선AD소서학습여기억적공능。
Objective To investigate the effects of Scutellaria baicalen-sis Georgi ( SBE ) on the activity of acetyl cholinesterase ( AChE ) and choline acetyltransferase ( ChAT) in hippocampus and cerebral cortex of the Alzheimer′s disease ( AD) model mice which induced by D-gal and NaNO2.Method AD model mice caused by D-gal (150 mg? kg -1 ) and NaNO2 (100 mg? kg-1 ) were intragastric administrad with 125, 250 and 500 mg? kg-1? d -1 SBE for 60 d.In 60 d after administration, mice were beheaded put to death,the protein content in cortex and hippo-campus were determinate, and the effect of SBE on the activity of AChE and ChAT in hippocampus and cerebral cortex were examined by Kits. Result The activity of AChE was higher and ChAT was lower in hippo-campus and cerebral cortex of the AD model mice than in the normal group.While in the AD model mice treated with SBE 500 mg? kg-1? d-1 , the activity of AChE in the hippocampus was significantly decreased, on the other hand, the activity of ChAT in the hippocampus was significantly increased.Treated with SBE 250,500 mg? kg-1? d-1 , the activity of ChAT in the cerebral cortex was significantly enhanced. Conclusion These results indicated that SBE encourages the synthesis and inhibits the decomposition of choline enzyme in hippocampus and cerebral cortex of the AD model mice.
