目的:观察抗CD20单克隆抗体(Anti-CD20)与白细胞介素10(IL-10)联合应用对非肥胖型糖尿病(NOD)小鼠脾脏免疫功能的影响。方法将24只NOD小鼠随机分为A、B、C、D组各6只,分别于第1、8、15、21天尾静脉注射Anti-CD20250μg、Anti-CD20250μg +IL-100.1 mL、IL-100.1 mL和生理盐水0.1 mL。首次用药后第12周断颈处死小鼠,立即取出脾脏,采用免疫组化法检测脾脏组织中的CD4+、CD3+T细胞及IL-27、IL-10。结果与D组比较,A、B、C组脾脏组织中CD4+、CD3+T细胞数量及IL-27、IL-10表达增加( P均<0.01);与A、C组比较,B组脾脏组织中CD+4、CD+3 T细胞数量及IL-27、IL-10表达增加(P均<0.01)。结论 Anti-CD20与IL-10联合应用可增加NOD小鼠脾脏组织中CD+4、CD+3 T细胞数量及IL-27、IL-10表达,从而调节机体免疫微环境,预防糖尿病的发生。
目的:觀察抗CD20單剋隆抗體(Anti-CD20)與白細胞介素10(IL-10)聯閤應用對非肥胖型糖尿病(NOD)小鼠脾髒免疫功能的影響。方法將24隻NOD小鼠隨機分為A、B、C、D組各6隻,分彆于第1、8、15、21天尾靜脈註射Anti-CD20250μg、Anti-CD20250μg +IL-100.1 mL、IL-100.1 mL和生理鹽水0.1 mL。首次用藥後第12週斷頸處死小鼠,立即取齣脾髒,採用免疫組化法檢測脾髒組織中的CD4+、CD3+T細胞及IL-27、IL-10。結果與D組比較,A、B、C組脾髒組織中CD4+、CD3+T細胞數量及IL-27、IL-10錶達增加( P均<0.01);與A、C組比較,B組脾髒組織中CD+4、CD+3 T細胞數量及IL-27、IL-10錶達增加(P均<0.01)。結論 Anti-CD20與IL-10聯閤應用可增加NOD小鼠脾髒組織中CD+4、CD+3 T細胞數量及IL-27、IL-10錶達,從而調節機體免疫微環境,預防糖尿病的髮生。
목적:관찰항CD20단극륭항체(Anti-CD20)여백세포개소10(IL-10)연합응용대비비반형당뇨병(NOD)소서비장면역공능적영향。방법장24지NOD소서수궤분위A、B、C、D조각6지,분별우제1、8、15、21천미정맥주사Anti-CD20250μg、Anti-CD20250μg +IL-100.1 mL、IL-100.1 mL화생리염수0.1 mL。수차용약후제12주단경처사소서,립즉취출비장,채용면역조화법검측비장조직중적CD4+、CD3+T세포급IL-27、IL-10。결과여D조비교,A、B、C조비장조직중CD4+、CD3+T세포수량급IL-27、IL-10표체증가( P균<0.01);여A、C조비교,B조비장조직중CD+4、CD+3 T세포수량급IL-27、IL-10표체증가(P균<0.01)。결론 Anti-CD20여IL-10연합응용가증가NOD소서비장조직중CD+4、CD+3 T세포수량급IL-27、IL-10표체,종이조절궤체면역미배경,예방당뇨병적발생。
Objective To observe the influence of anti-CD20 monoclonal antibody ( Anti-CD20 ) combined with inter-leukin 10 ( IL-10) on the immune function of spleen in non-obese diabetic ( NOD) mouse.Methods Twenty-four NOD mice were randomly divided into four groups , 6 mice in each group:groups A, B, C and D, which were caudal venously injected with 250μg Anti-CD20 , 250μg Anti-CD20 +0.1 mL IL-10, 0.1 mL IL-10 and 0.1 mL normal saline at day 1, 8, 15 and 21, respectively.All mice were sacrificed 12 weeks after first injection and the spleens were immediately taken out . The number of CD4+T cells, CD3+T cells and the levels of IL-10 and IL-27 were detected by immunohistochemistry . Results Compared with group D, the number of CD+4 T cells, CD+3 T cells and the levels of IL-10 and IL-27 in the spleen tissues were increased in the groups A , B and C (all P<0.01).Compared with groups A and C , the number of CD4+T cells, CD+3 T cells and the levels of IL-10 and IL-27 in the spleen tissues were increased in the group B (all P<0.01). Conclusion The combined administration of Anti-CD20 and IL-10 increases the number of CD+4 T cells, CD+3 T cells and the levels of IL-10 and IL-27 in the spleen tissues , and thus it regulates the immune microenvironment and prevents the oc-currence of diabetes .
