目的:观察人脐静脉内皮细胞( HuVECs )分泌物对自身迁移和侵袭的影响。方法将传代的HuVECs分为3组,A、B组分别加入一定浓度的FBS及HuVECs培养24、48 h的上清液,C组加一定浓度的FBS及纯DMEM培养液。采用细胞划痕实验观察细胞迁移距离,采用 Transwell 迁移和侵袭实验观察细胞迁移和侵袭个数。结果 A、B、C组迁移距离分别为(219.27±28.44)、(163.64±19.67)、(158.18±16.36)μm;A组与B、C组比较, P均<0.05;B、C组比较,P>0.05。 A组迁移细胞数多于B、C组(P均<0.01),B、C组迁移细胞数无明显差异(P>0.05);A组侵袭细胞数少于B组,B组少于C组,P均<0.01。结论 HuVECs分泌物可在一定程度上促进细胞迁移、抑制细胞侵袭。

목적:관찰인제정맥내피세포( HuVECs )분비물대자신천이화침습적영향。방법장전대적HuVECs분위3조,A、B조분별가입일정농도적FBS급HuVECs배양24、48 h적상청액,C조가일정농도적FBS급순DMEM배양액。채용세포화흔실험관찰세포천이거리,채용 Transwell 천이화침습실험관찰세포천이화침습개수。결과 A、B、C조천이거리분별위(219.27±28.44)、(163.64±19.67)、(158.18±16.36)μm;A조여B、C조비교, P균<0.05;B、C조비교,P>0.05。 A조천이세포수다우B、C조(P균<0.01),B、C조천이세포수무명현차이(P>0.05);A조침습세포수소우B조,B조소우C조,P균<0.01。결론 HuVECs분비물가재일정정도상촉진세포천이、억제세포침습。
Objective To observe the influence of the secretion of human umbilical vein endothelial cells (HuVECs) on their own migration and invasion .Methods The passage HuVECs were divided into 3 groups: groups A, B and C. Groups A and B were added with a certain concentration of FBS and HuVECs supernatant with 24-hour and 48-hour cultiva-tion, meanwhile, group C was added with a certain concentration of FBS and pure DMEM culture solution .Cell scratch as-say was used to observe cell migration distance .The number of cell migration and invasion of HUVECs was observed with Transwell.Results The migration distance of groups A, B and C was (219.27 ±28.44), (163.64 ±19.67) and (158.18 ±16.36) μm, respectively.Significant difference was found between group A and groups B and C ( all P<0.05);and no significant difference was found between group A and group B (P>0.05).The migrated cells in the group A were more than those of groups B and C (all P<0.01), and no significant difference in the migrated cells was found be-tween group B and group C (all P>0.05).The invasive cells of group A were less than those of group B , and the invasive cells of group B were less than those of group C (all P<0.01).Conclusion The secretion of HUVECs may promote the cell migration and inhibit the cell invasion to a certain extent .
