目的：：探讨不同激活方法小鼠卵母细胞孤雌激活的效率及纺锤体的变化。方法：小鼠卵母细胞采用乙醇、氯化锶化学激活，研究孤雌激活效果；采用免疫荧光染色法观察孤雌激活下卵母细胞纺锤体的动态变化。结果：(1)8%乙醇处理10、15 min的桑囊率高于处理5 min；(2)10 mmol·L-1氯化锶激活率高于2.5、5 mmol·L-1；(3)激活过程中纺锤体形态发生了改变。结论：8%乙醇处理时间相对延长有利于提高桑囊率；高浓度氯化锶激活效果较好；孤雌激活后纺锤体形态会发生改变。
목적：：탐토불동격활방법소서란모세포고자격활적효솔급방추체적변화。방법：소서란모세포채용을순、록화송화학격활，연구고자격활효과；채용면역형광염색법관찰고자격활하란모세포방추체적동태변화。결과：(1)8%을순처리10、15 min적상낭솔고우처리5 min；(2)10 mmol·L-1록화송격활솔고우2.5、5 mmol·L-1；(3)격활과정중방추체형태발생료개변。결론：8%을순처리시간상대연장유리우제고상낭솔；고농도록화송격활효과교호；고자격활후방추체형태회발생개변。
Objective:To evaluate the efficiency of mice oocytes parthenogenetic activation by different activation methods and the change of the spindle.Method:Oocytes were obtained from mice ovaries.Parthenogenetic activation effect of mouse oocytes were re-searched after activation by ethanol,strontium chloride chemical activation.Oocytes dynamic changes of the spindle were observed by immunofluorescence staining after parthenogenetic activation.Results:(1)Mulberry sac rate of 8% ethanol processing 10,15 min is higher than the treatment 5 min;(2)the strontium chloride activation rate for 10 mmol·L-1 is higher than 2.5,5 mmol·L-1;(3) Spindle shape has changed during the process of activation.Conclusion:Relative to extend the processing time of 8% ethanol is bene-ficial to increase mulberry sac rate;High concentrations of strontium chloride activation effect is better;spindle morphology changes after parthenogenetic activation.
