目的::观察积雪草苷对人胃癌 SGC-7901细胞株转移能力和凋亡的影响。方法:采用细胞计数法(CCK-8)检测不同浓度的积雪草苷对人胃癌 SGC-7901细胞株增殖影响;细胞黏附试验检测细胞体外黏附能力的变化,Transwell 试验检测细胞体外侵袭能力的变化,流式细胞仪 AnnexinV/PI双染法检测细胞凋亡率。结果:积雪草苷明显抑制人胃癌 SGC-7901细胞株增殖,积雪草苷处理后人胃癌 SGC-7901细胞株黏附能力分别下降到61.2%,47.6%,38.6%;侵袭能力分别下降到42.3%,31.7%,21.6%;AnnexinV/PI双染法检测显示积雪草苷可诱导人胃癌 SGC-7901细胞株凋亡,与阴性对照组比较,差异有统计学意义(P<0.01)。结论:积雪草苷能抑制人胃癌 SGC-7901细胞株的增殖,抑制其体外粘附和侵袭能力,诱导其凋亡。

목적::관찰적설초감대인위암 SGC-7901세포주전이능력화조망적영향。방법:채용세포계수법(CCK-8)검측불동농도적적설초감대인위암 SGC-7901세포주증식영향;세포점부시험검측세포체외점부능력적변화,Transwell 시험검측세포체외침습능력적변화,류식세포의 AnnexinV/PI쌍염법검측세포조망솔。결과:적설초감명현억제인위암 SGC-7901세포주증식,적설초감처리후인위암 SGC-7901세포주점부능력분별하강도61.2%,47.6%,38.6%;침습능력분별하강도42.3%,31.7%,21.6%;AnnexinV/PI쌍염법검측현시적설초감가유도인위암 SGC-7901세포주조망,여음성대조조비교,차이유통계학의의(P<0.01)。결론:적설초감능억제인위암 SGC-7901세포주적증식,억제기체외점부화침습능력,유도기조망。
Objective:To investigate the effects of asiaticoside on proliferation and apoptosis of SGC-7901.Methods:Growth inhi-bition was measured by cell counting kit-8,the adhesion in vitro was detected by cell adhesion assay;the invasion in vitro was detec-ted by transwell assay;the rate of apoptosis was detected by flow cytometry with Annexin-V/PI staining.Results:Asiaticoside inhib-ited the growth of SGC-7901 effectively.The adhesion of cells in vitro was decreased to 61.2% and 38.6% respectively;the invasion in vitro was decreased to 42.3% and 21.6%.Compared with the negative control group,the apoptosis of SGC-7901was significantly induced by different concentration of asiaticoside (P<0.01).Conclusion:Asiaticoside can inhibit the proliferation,induce apoptosis of SGC-7901 and inhibit the metastasis of SGC-7901 in vitro.
