目的:通过复方地芬诺酯法建立大鼠慢传输型便秘模型,探索一种简便易行的大鼠便秘造模方法,并探讨大鼠便秘造模过程中对结肠AQP8表达的影响。方法:SD大鼠30只随机分为空白组和便秘模型组。空白组大鼠用生理盐水灌胃,便秘模型组用复方地芬诺酯灌胃,观察两组大鼠大便粒数及大便湿重。大鼠便秘模型建立后。将大鼠集体处死,取大鼠近端结肠标本,用免疫组织化学的方法检测两组大鼠结肠AQP8的表达情况。使用彩色病理图像分析系统对阳性表达的平均光密度值进行半定量分析。结果:建立便秘模型末次给药后,便秘组24 h大便粒数、大便湿重均低于空白对照组(P<0.05),差异有统计学意义,表明大鼠慢传输型便秘模型成功建立;免疫组化结果显示:便秘组结肠AQP8的MD值大于空白对照组(P<0.05),差异有统计学意义。结论:复方地芬诺酯灌胃法成功建立大鼠便秘模型,便秘模型的建立与大鼠结肠AQP8的表达增高相关。
目的:通過複方地芬諾酯法建立大鼠慢傳輸型便祕模型,探索一種簡便易行的大鼠便祕造模方法,併探討大鼠便祕造模過程中對結腸AQP8錶達的影響。方法:SD大鼠30隻隨機分為空白組和便祕模型組。空白組大鼠用生理鹽水灌胃,便祕模型組用複方地芬諾酯灌胃,觀察兩組大鼠大便粒數及大便濕重。大鼠便祕模型建立後。將大鼠集體處死,取大鼠近耑結腸標本,用免疫組織化學的方法檢測兩組大鼠結腸AQP8的錶達情況。使用綵色病理圖像分析繫統對暘性錶達的平均光密度值進行半定量分析。結果:建立便祕模型末次給藥後,便祕組24 h大便粒數、大便濕重均低于空白對照組(P<0.05),差異有統計學意義,錶明大鼠慢傳輸型便祕模型成功建立;免疫組化結果顯示:便祕組結腸AQP8的MD值大于空白對照組(P<0.05),差異有統計學意義。結論:複方地芬諾酯灌胃法成功建立大鼠便祕模型,便祕模型的建立與大鼠結腸AQP8的錶達增高相關。
목적:통과복방지분낙지법건립대서만전수형편비모형,탐색일충간편역행적대서편비조모방법,병탐토대서편비조모과정중대결장AQP8표체적영향。방법:SD대서30지수궤분위공백조화편비모형조。공백조대서용생리염수관위,편비모형조용복방지분낙지관위,관찰량조대서대편립수급대편습중。대서편비모형건립후。장대서집체처사,취대서근단결장표본,용면역조직화학적방법검측량조대서결장AQP8적표체정황。사용채색병리도상분석계통대양성표체적평균광밀도치진행반정량분석。결과:건립편비모형말차급약후,편비조24 h대편립수、대편습중균저우공백대조조(P<0.05),차이유통계학의의,표명대서만전수형편비모형성공건립;면역조화결과현시:편비조결장AQP8적MD치대우공백대조조(P<0.05),차이유통계학의의。결론:복방지분낙지관위법성공건립대서편비모형,편비모형적건립여대서결장AQP8적표체증고상관。
Objective:to establish a rat model of slow transit constipation by compound diphenoxylate, to explore a simple method of the establishment of constipation model, and discuss the influence on the expression of AQP8 in colon of rat in modeling process. Methods:30 SD rats were randomly divided into blank group and constipation model group.The rats of blank group were given normal saline by gastric irrigation,while the rats of model group were given Diphenoxylate by intragastric administration.To observe the stool grains and stool wet weight of the rats of two groups.After establishment the rat model of constipation, killed the rats collective and collected the rats, proximal colons.To detect expression of AQP8 in the proximal colon by immunohistochemistry.The staining results were analyzed semi-quantitively by computerized color image analyzer using the following parameter:mean density. Results: After the last administration to establish the model of STC, both of stool wet weight and grain number of the blank control group were higher than the other four groups, (P<0.05), the difference was statistically significant, which show that we had established the rat model of slow transit constipation successfully;immunohistochemistry results showed that:Immunohistochemical results:The density mean(MD) of AQP8 in the colon of model group was greater than the blank control group (P<0.05), the difference was statistically significant. Conclusion:The rat model of constipation was successfully established by compound diphenoxylate intragastric administration,it had correlation between the establishment of constipation model and the increased expression of AQP8 in colon of rats.
