为从藏药中发现新的抗癌药物,从青海互助县收集了100种常用藏药,采用快速溶剂萃取法制备出了各种藏药的环己烷、乙酸乙酯和甲醇提取物。选用2种肝癌细胞株HepG2和SMMC-7721,以MTT法测试了100种藏药的抗肝癌活性,选用人肝细胞株LO-2评价了藏药的体外毒性。结果表明:15种藏药材显示出抗肝癌活性,IC50值均小于150μg/mL,其中藜、天名精、三脉紫菀3种藏药的乙酸乙酯提取物显示出显著的抗肝癌活性,其IC50值均小于50μg/mL,而对正常肝细胞LO-2未显示出明显的毒性。该研究为进一步从藏药中发现新的抗癌药物提供了思路。

위종장약중발현신적항암약물,종청해호조현수집료100충상용장약,채용쾌속용제췌취법제비출료각충장약적배기완、을산을지화갑순제취물。선용2충간암세포주HepG2화SMMC-7721,이MTT법측시료100충장약적항간암활성,선용인간세포주LO-2평개료장약적체외독성。결과표명:15충장약재현시출항간암활성,IC50치균소우150μg/mL,기중려、천명정、삼맥자완3충장약적을산을지제취물현시출현저적항간암활성,기IC50치균소우50μg/mL,이대정상간세포LO-2미현시출명현적독성。해연구위진일보종장약중발현신적항암약물제공료사로。
To discover new anticancer drugs from Tibetan medicines, the cyclohexane, ethyl acetate and methanol extracts from 100 commonly used Tibetan medicines collected from Huzhu County, Qinghai Province were prepared by an accelerated solvent extraction method.The MTT method was used to test the anti-hepatoma activity of the 100 Tibetan medicines against two hepatocellular carcinoma cell lines HepG2 and SMMC-7721, and the preliminary cell toxicity was investigated using a human liver cell line LO-2.The results indicated that 15 of these Tibetan medicines exhibited anti-hepatoma activities against HepG2 and SMMC-7721 with IC50 values less than 150μg/mL.Among these, the ethyl acetate extracts of Delphinium caeruleum, Dracocephalum tanguticum and Ligularia przewalskii showed significant antitumor activity with IC50 values less than 50 μg/mL, yet they didn’ t show obvious toxicity against LO-2 cells.This research presented a new way to discover new anti-hepatoma drugs from Tibetan medicines.
