目的:观察补肾调肝方对膝骨关节炎模型大鼠软骨下骨BMCs骨保护素(OPG)的影响。方法:5月龄清洁级健康雌性SD大鼠30只随机分为Sham组、模型组、中药组,造模成功后给药,中药组术后第4周开始灌服补肾调肝方,模型组、sham组灌服等量生理盐水,按期分批处死,取软骨下骨BMSCs培养,用免疫组化方法检测BMSCs的OPG表达水平。结果:中药组OPG表达阳性细胞百分比均显著高于Sham组及模型组(P<0.01)。结论:补肾调肝方可升高OPG,阻止或改善大鼠膝关节软骨降解和退变,达到防治骨关节炎的目的。

목적:관찰보신조간방대슬골관절염모형대서연골하골BMCs골보호소(OPG)적영향。방법:5월령청길급건강자성SD대서30지수궤분위Sham조、모형조、중약조,조모성공후급약,중약조술후제4주개시관복보신조간방,모형조、sham조관복등량생리염수,안기분비처사,취연골하골BMSCs배양,용면역조화방법검측BMSCs적OPG표체수평。결과:중약조OPG표체양성세포백분비균현저고우Sham조급모형조(P<0.01)。결론:보신조간방가승고OPG,조지혹개선대서슬관절연골강해화퇴변,체도방치골관절염적목적。
Objective:To observe the effect of tonifying liver and kidney decoction on BMSCs OPG expression of SD rats with knee osteoarthritis subchondral bone. Methods:30 female clean and healthy SD rats of 5 months old were randomly divided into sham group, model group and Chinese medicine group, administered after successful modeling, Chinese medicine group began intragastric administration of tonifying liver and kidney de-coction in fourth week postoperatively, model group and sham group with gavage saline of the same amount, regularly executed in turn, subchondral bone BMSCs was cultured, the expression level of BMSCs OPG was detected with immunohistochemical method. Results:The percentage of OPG positive cells in Chinese medicine group was both significantly higher than that in Sham group and model group (P<0.01). Conclusion:Tonifying liv-er and kidney decoction can increase OPG, prevent or improve the degradation and degeneration of knee joint cartilage of rats, to achieve the purpose of preventing and treating osteoarthritis.
