目的 探讨糖尿病肾病(diabetic nephropathy,DN)患者血浆差异蛋白表达情况,为寻找DN敏感血浆蛋白标志物提供科学依据.方法 对6例DN患者的血浆与6例正常人血浆应用双向荧光差异凝胶电泳技术(2D-DIGE)观察疾病蛋白质标记物,选择表达差异大于1.5倍的蛋白斑点进行质谱分析.结果 成功建立糖尿病肾病和正常血浆的胶内差异双向凝胶电泳图谱,通过分析共鉴定出1 1种差异蛋白质,包括C3补体、C4补体、载脂蛋白E等.结论 2D-DIGE能够客观地显示DN患者血浆与正常血浆之间的蛋白质表达差异,本研究鉴定的11种蛋白有可能为DN患者血浆蛋白分子标志物,可为DN的早期诊断提供依据.

Objective To investigate the expression of plasma proteins in patients with diabetic nephropathy (DN),to provide a scientific basis for searching for sensitive plasma protein markers in DN.Methods 6 patients with DN and 6 normal subjects were observed by two-dimensional fluorescence difference gel electrophoresis (2D-DIGE) in plasma to study disease protein markers.Selected protein spots with differential expression was greater than 1.5 fold to give mass spectrometry analysis.Results A two-dimensional gel electrophoresis profile of diabetic nephropathy and normal plasma was successfully established.11 different proteins were identified by analysis,including C3,C4,Apolipoprotein E,etc.Conclusion 2D-DIGE can objectively show the difference of protein expression between DN patients and normal blood plasma.11 proteins identified in this study may be molecular markers for DN,can provide the basis for the early diagnosis of DN.
