目的：建立高效液相色谱法（HPLC）测定尿样中美沙酮浓度的方法。方法：Waters C18 ODS色谱柱（3．9mm ×150mm，5μm），流动相为乙腈-磷酸二氢钾（32∶68），流速1mL·min-1检测波长为220nm。盐酸苯海索为内标。结果：方法回收率为99．1％～107．17％，日内相对标准偏差RSD为1．16％～3．52％，日间RSD为1．14％～3．58％，线性范围为200～10000ng·mL-1。结论：本法简便、准确、可重复性好，可用于尿样中美沙酮浓度的测定。
목적：건립고효액상색보법（HPLC）측정뇨양중미사동농도적방법。방법：Waters C18 ODS색보주（3．9mm ×150mm，5μm），류동상위을정-린산이경갑（32∶68），류속1mL·min-1검측파장위220nm。염산분해색위내표。결과：방법회수솔위99．1％～107．17％，일내상대표준편차RSD위1．16％～3．52％，일간RSD위1．14％～3．58％，선성범위위200～10000ng·mL-1。결론：본법간편、준학、가중복성호，가용우뇨양중미사동농도적측정。
Objective:To develop a HPLC method for the determination of methadone concentration in urine.Methods:Waters C18 ODS column(3.9mm ×150mm,5μm)was used.The mobile phase was acetonitrile-monopotassium phosphate (32∶68).The flow rate was 1 mL·min-1 and the UV detection wavelength was 220nm.The internal standard was trihexyphenidyl hydrochloride.Results:The recovery rate was 99.1%~107.17%.The within-day relative standard deviation (RSD)was 1.16%~3.52%,and between-day RSD was 1.14%~3.58%.It had a good linear relationship within 200~10000ng·mL-1.Conclusion:The method is simple,accurate and reproducible,and can be used for drug determination of methadone in urine.
