目的:探讨TGF-β1和bFGF在膝骨关节炎患者骨赘中和关节软骨表达和分布的情况。方法:取12例诊断为膝骨关节炎且在影像学上可有明显的骨赘形成并行全膝置换患者的膝关节软骨以及骨赘,采用免疫组化对TGF-β1和bFGF表达以及分布情况进行分析。结果:免疫组化链菌素亲生物素蛋白-过氧化酶连接法证实在12例标本的关节软骨和骨赘中TGF-β1和bFGF均有表达;TGF-β1在骨赘的浅层有高强度阳性表达,在深层未有表达;而在退变软骨的浅层只有弱强度阳性表达,深层为阴性表达;而且TGF-β1在骨赘中的表达强度要强于关节软骨。而bFGF在骨赘和软骨的整个层面均有阳性表达。结论:TGF-β1和bFGF参与了骨赘的形成和软骨退变,对骨关节炎的进展有着重要的影响,TGF-β1和bFGF这种表达的差异性,表明这2种生长因子在骨关节炎患者骨赘形成中对骨与软骨代谢起着不同的作用,TGF-β1相比bFGF似乎与骨赘的形成更加相关。

목적:탐토TGF-β1화bFGF재슬골관절염환자골췌중화관절연골표체화분포적정황。방법:취12례진단위슬골관절염차재영상학상가유명현적골췌형성병행전슬치환환자적슬관절연골이급골췌,채용면역조화대TGF-β1화bFGF표체이급분포정황진행분석。결과:면역조화련균소친생물소단백-과양화매련접법증실재12례표본적관절연골화골췌중TGF-β1화bFGF균유표체;TGF-β1재골췌적천층유고강도양성표체,재심층미유표체;이재퇴변연골적천층지유약강도양성표체,심층위음성표체;이차TGF-β1재골췌중적표체강도요강우관절연골。이bFGF재골췌화연골적정개층면균유양성표체。결론:TGF-β1화bFGF삼여료골췌적형성화연골퇴변,대골관절염적진전유착중요적영향,TGF-β1화bFGF저충표체적차이성,표명저2충생장인자재골관절염환자골췌형성중대골여연골대사기착불동적작용,TGF-β1상비bFGF사호여골췌적형성경가상관。
Objective:To explore the expression and localization of Transforming growth factor-β1 and Basic Fibroblast Growth Factor in the cartilage and osteophytes in knee osteoarthritis patients.Methods:Cartilage and osteophytes from 12 patients diagnosed with primary knee OA with obvious radiographic osteophytes were examined using immunohistochemistry.Results: Labeled streptavidin biotin method demonstrated that TGF-β1 and bFGF were detected in all 12 osteophytes and cartilage;TGF-β1 expression was highly positive in super-ficial area but almost negative in deep layer in osteophytes and weak in superficial area of cartilage,and levels of expression in osteophytes were higher than in cartilage .bFGF expression were all detected in the whole layer of both osteophyte and degenerative cartilage.Conclu-sion:TGF-β1 and bFGF were both involved in the progresstion of osteophytes formation and cartilage degeneration,which played great roles in OA development.The expression and localization difference between these two growth factors suggested their different roles in bone and cartilage metabolism in osteophyte formation,TGF-β1 seems to be more associate with the formation of osteophytes.
