目的:探讨阿胶对围绝经期大鼠卵巢颗粒细胞凋亡及其相关基因Bcl-2和Bax表达的影响。方法:选用4月龄雌性大鼠为正常对照组,14月龄阴道细胞学表现为动情期延长的雌性大鼠为衰老大鼠模型。模型动物随机分别给予阿胶或戊酸雌二醇片治疗。采用原位末端核苷酸标记法(TUNEL)检测卵巢细胞凋亡,免疫组化法检测细胞凋亡相关因子Bcl-2,Bax。结果:阿胶高、中、低剂量组卵巢细胞凋亡阳性表达与模型组比较有显著性差异,同时可使卵巢凋亡基因Blc-2表达增强,促凋亡基因Bax表达减弱,Bcl-2/Bax比例增加。结论:阿胶通过抑制Bax蛋白的表达,上调Bcl-2蛋白表达,抑制卵巢颗粒细胞凋亡,进而改善卵巢功能。
目的:探討阿膠對圍絕經期大鼠卵巢顆粒細胞凋亡及其相關基因Bcl-2和Bax錶達的影響。方法:選用4月齡雌性大鼠為正常對照組,14月齡陰道細胞學錶現為動情期延長的雌性大鼠為衰老大鼠模型。模型動物隨機分彆給予阿膠或戊痠雌二醇片治療。採用原位末耑覈苷痠標記法(TUNEL)檢測卵巢細胞凋亡,免疫組化法檢測細胞凋亡相關因子Bcl-2,Bax。結果:阿膠高、中、低劑量組卵巢細胞凋亡暘性錶達與模型組比較有顯著性差異,同時可使卵巢凋亡基因Blc-2錶達增彊,促凋亡基因Bax錶達減弱,Bcl-2/Bax比例增加。結論:阿膠通過抑製Bax蛋白的錶達,上調Bcl-2蛋白錶達,抑製卵巢顆粒細胞凋亡,進而改善卵巢功能。
목적:탐토아효대위절경기대서란소과립세포조망급기상관기인Bcl-2화Bax표체적영향。방법:선용4월령자성대서위정상대조조,14월령음도세포학표현위동정기연장적자성대서위쇠로대서모형。모형동물수궤분별급여아효혹무산자이순편치료。채용원위말단핵감산표기법(TUNEL)검측란소세포조망,면역조화법검측세포조망상관인자Bcl-2,Bax。결과:아효고、중、저제량조란소세포조망양성표체여모형조비교유현저성차이,동시가사란소조망기인Blc-2표체증강,촉조망기인Bax표체감약,Bcl-2/Bax비례증가。결론:아효통과억제Bax단백적표체,상조Bcl-2단백표체,억제란소과립세포조망,진이개선란소공능。
Objective:To study the effects of Colla Corii Asini on ovarian granulosa cells apoptosis and expression of Bcl-2 and Bax in perimenopausal rats.Methods:SD female rats of 4 months old were set to be the normal control group and SD female rats of 14 months old whose oestrus elongation in colpocytology appearance were set as the senile female rat model.These rats were divided into 5 groups randomly, as follows:high dose Colla Corii Asini group, middle dose Colla Corii Asini group, low dose Colla Corii Asini group, estradiol valerate group, model group.After 8 weeks in the intervention period, ovarian apoptosis and the expression of ovarian granulosa cell Bcl-2, Bax were measured by TUNEL and immunohisto-chemistry methods.Results:High and middle dose Colla Corii Asini groups could de-crease apoptosis in ovarian granulose cells (P<0.05), and could improve the expression of Bcl-2 in the ovaries and weakened the ex-pression of Bax, which then increase the ratio of Bcl-2/Bax.Conclusion:Colla Corii Asini could delay ovarian granulose cells apoptosis through inhibiting the expression of Bax, increasing the expression of Bcl-2 during perimenopausal period, which might be one reason of Colla Corii Asini postponed the ovarian failure and cured perimenopausal syndrome.
